The Development Of Qizhu Rhinitis Granules

Objective:To improve allergic rhinitis compound preparation of traditional Chinese medicines "Qizhu rhinitis granules",and study its production process,quality standard,pharmacodynamics. Methods:According to properties of various kinds of medicinal materials in prescription, the extraction used fractionation technology and granular molding adoped fluidization spray granulating process.Magnoliabark, atractylodes,citrus used ethanol extraction process that were optimized using the method of the single factor investigation, the orthogonal test, the ethanol concentration and recovery. Astragalus adopted reflux extraction process that were optimized using the method of the orthogonal experiment,the ethanol precipitation and enrichment process. Poria used water extraction process that were optimized using the method of the single factor investigation. Determined the best granular molding process using the single factor investigation method.Established the allergic rat model that ovalbumin induced, observed the influence of Qizhu rhinitis granules on the symptom of allergic rhinitis and serum IgE level. Results:With the single factor investigation method, magnolia bark, atractylodes, citrus used alcohol extraction process adaptively.The result of orthogonal experiment showed that the best extraction process were 60% ethanol extraction three times, adding 8 times,6 times,4 times resolver respectively,1.5 hours every time decoction. Through the orthogonal experiment, the best reflux extraction conditions of astragalus membranaceus were:medicinal materials soaking for 10 min, heating reflux extraction two times,1.5 hours every time, adding 4 times,2 times resolver respectively. Using 40% alcohol content conditions removed impurities. Astragalus membranaceus alcohol fluid were combined with the alcohol extract fluid of atractylodes, citrus; The mixed liquids were concentrated with the conditions of 70 ℃ to 75 ℃, and 0.06-0.08 Mpa, the active ingredients are retained well. Poria cocos with water decoction extraction, the optimum extraction process through the single factor investigation concluded:adding six times water, decocting 2 times,2 hours each time, filtered, vacuum concentration until the concentration of fluid of 1 g/ml(crude drug) with 70 ℃ to 75 ℃,0.06-0.08 Mpa. Merged the above clear cream, concentrated to the relative density of 1.05-1.10 (60 ℃). Added right amount of the dextrin, powdered sugar, blended, fluidified spray granules to 1000 g. The quality control determined the indexs of magnolol, honokiol,astragaloside:there were 0.60 mg magnolol and honokiol, not less than 0.10 mg astragaloside per 1 g granules. In qualitative identification, the spots of citrus, atractylodes were clear, and not other distractions.Qizhu rhinitis granule could obviously improve the symptoms of allergic rhinitis, reduce serum total IgE level. Conclusion:Preparation process of Qizhu rhinitis granule was stability, quality was controlled and inspection method was accurate, and had good inhibition, treatment effect of allergic rhinitis.
