clinical research.method:1.Preparation process:Inspect the factors such as the amount of water added,the number of cooking,and the time of boiling.The dry extract,total saponins of Astragalus and sinapine thiocyanate content were optimized for the test;The effect of Shutong digesting biscuits was optimized by the concentration of sinapine thiocyanate transfer rate;According to factors such as the temperature of the inlet and outlet air,the spray drying process is optimized with the amount of dry paste powder,moisture content and dry paste yield.2.Quality standard:Refer to the 2015 edition of the Chinese Pharmacopoeia for research and verification,Formulate physical and chemical indicators such as moisture,ash,alkalinity,acid value,and peroxide value;Astragalus and radish seed selection and quality control of qualitative indicators,establishing TLC methods;Selecting the main index component of radish seed sinapine thiocyanate as a quantitative quality control index,establishing its high performance liquid chromatography method.3.Pharmacodynamic:Selecting loperamide hydrochloride to establish the constipation model of ICR mice,using mosapride citrate as a positive control drug.Through the determination of mice's ink promotion rate in the small intestine,we explored the effect of this Shutong digestive biscuit extract on experimental constipation.The effect of murine intestinal motility;By observing the fecal traits and the time of first row of black feces,the effects of the Shutong digestive biscuit extract on the bowel movement of experimental constipation ICR mice were investigated.Result:1.The optimum extraction and preparation process of the Shutong digestive biscuits : 8 times water,boiled twice,each boiling 90 min;extract concentration temperature: select 80 °C and below for decompression concentration;outlet temperature: 102 °C The relative density of the dry extract was 1.25g/ml,and spray drying was carried out without the aid of additives at the inlet air temperature of 180°C.2.The physical and chemical indicators of Shutong digestive biscuits are in line with the standards,and the quality is stable.In the thin layer chromatography identification of radish seed and Radix Astragali through Shutong digestive biscuits,the chromatogram of the sample is in the same position as the chromatogram of the reference medicinal material,showing the same color.The main fluorescent spot,and the negative sample in the corresponding position without spots;sinapine thiocyanate was determined by high performance liquid chromatography,the linear regression equation was Y = 14693X-3696.2(r2 = 0.9999),The linear relationship was good in the range of 3.03 ~ 45.42?g/ml;the precision RSD was 1.10%,the recovery of the sample was 98.75%,and the RSD was 2.04%;the sample solution was stable within 12 hours and no negative interference was observed.The reproducibility RSD was 0.827%.3.Compared with the control group,the mice in the model group had a significant difference in the first row of melena time and intestinal ink promotion rate(p <0.01).Compared with the mice in the model group,the time for the first melena in the mice in the middle-dose group decreased,and there was a significant difference(p <0.05).The time for the first row ofmelena in the high-dose group was significantly reduced,with significant difference(p <0.01);Compared with the mice in the model group,The middle-dose group mice's ink-propulsion rate of the small intestine was increased,with significant differences(p <0.05).The high-dose group mice's ink-propulsion rate of the small intestine was significantly increased,with significant differences(p <0.01).Conclusion: The preparation technology of the Shutong digesting biscuits is reasonable,the quality standard is stable,and the stability is good.The Shutong digestive biscuit extract has a good laxative effect on the constipation mice,and the laxative effect has a dose-effect relationship with the dose of the test substance,and has further research and development value.
