Cyclodextrin (CD), as one of the most important supramolecular compounds, with its unique molecular structure and physical and chemical properties, has been the focus of scientists’ research in recent decades. CD plays an important role in pharmaceutical excipients, drug carrier, organic catalysis, molecular devices, analysis, testing and chromatographic study. Moreover, the drug excipients and drug carrier are the most valuable application. The most frequently used modified procedures in the field of drug research are two ways:1) as a kind of drug excipient, CDs can from to inclusion complexs with poor water soluble or low bioavailability of organic small molecule drugs to improve the water solubility and bioavailability of them.2) as a kind of drug carrier, through chemical methods, the drug molecules can be gifted to the cyclodextrin molecule to improve the water soluble and target of drug molecules. Therefore, in order to further expand the application of cyclodextrin in medicinal chemistry and research the molecular inclusion mechanism of cyclodextrins and guest, in this paper we selected daidzin as guest to research the incusion behavior with cyclodextrins. First of all, we select the HP-β-CD and SBE-P-CD which have been approved by the FDA to form inclusion complexes with daidzein.Then the inclusion mode and solubilization have been further researched.This dissertation was mainly constituted of three sections as follows:1. The preparation of β-cyclodextrin amine derivatives:Four amino modified β-cyclodextrin derivatives were obtained:mono-6-(amino)-6-deoxy-β-cyclodextrin (NH2-β-CD), mono-6-(ethylene-diamino)-6-deoxy-β-cyclodextrin (EN-P-CD), mono-6-(diethylenetriamine)-6-deoxy-β-cyclodextrin (DETA-β-CD), mono-6-(triethylene tetramine)-6-deoxy-β-cyclodextrin (TETA-β-CD) and all of their structures were charactered by lH NMR.2. The preparation of daidzein/HP-β-CD and SBE-β-CD inclusion complexs:HP-β-CD and SBE-P-CD are selected as host and the inclusion complexes were prepared in a green method. Fluorescence spectrometry and Job curve methods were used to obtain the inclusion ratio and the constant Ks.1H NMR and 2D NMR Roesy were amployed to research the possible inclusion mode. In addition, XRD, TG,DSC, SEM were also used to identify the formation of inclusion complexes.3. The preparation of daidzein/NH2-β-CD and EN-β-CD inclusion complexs:HP-β-CD and SBE-β-CD are selected as host to get the new inclusion complexes with daidzein. Fluorescence spectrometry, Job curve, NMR, XRD, TG, DSC, SEM were used to identify the formation of inclusion complexes.
