Ginsenosides are triterpenes saponins considered to be the main bioactive principles of the most important Oriental herbal medicine "ginseng" derived from the roots and rhizomes of different Panax species (Araliaceae). The most commonly used Panax species are P. ginseng (Korean orAsian ginseng), P. quinquefolius (American ginseng), P. notoginseng (Tienchi or Sanchi). Ginsenosides are considered to promote longevity,especially for increase resistance to decrepit,potential cancer preventive effects, bioactivities for human cardiovascular system.A huge quantity of work has been carried out in order to isolate and prepare the ginsenosides, but few research of the flow discipline has been carried. In order to study the flow discipline, ginseng triterpenes, quinquefolius leaf triterpenes, notoginseng triterpenes, notoginseng leaf triterpenes were performed in 8 common solvent system. Then isolation and identification of the ginsenosides from quinquefolius leaf triterpenes were researched.In the article, TLC was the technique for the ginsenosides analysis, ginseng triterpenes, quinquefolius leaf triterpenes,notoginseng triterpenes, notoginseng leaf triterpenes were performed in 8 common solvent system. The results indicated that the mixture of chloroform, methanol and water (65:35:10) solvent system is the best for the ginseng triterpenes in all solvent system; the mixture of n-butyl alcohol, ethyl acetate and water (40:10:50) solvent system is the best for the quinquefolius leaf triterpenes in all; the mixture of chloroform, methanol and water (65:35:10) solvent system is the best for notoginseng triterpenes in all; the mixture of n-butyl alcohol, ethyl acetate water and ethylether(40:10:5:1) solvent system is the best for notoginseng leaf triterpenes in all. we can see the mixture of chloroform, methanol and water (65:35:10) solvent system is fit to separate ginsenosides.Eight ginsenosides were isolated by vacuum liquid chromatography and SG-64 chromatography from P. quinquefolius. Their structures were identified as ginsenosides Rgi(Ⅰ), ginsenosides Re(Ⅵ), ginsenosides Rd (Ⅷ), ginsenosides La/Rg3 (Ⅱ) ginsenosides F2 (Ⅴ), Pseudo-ginsenoside Fa (Ⅶ), respectively. Compounds Ⅲ,Ⅳ were ginsenosides Rg2 and unknown.
