2-(N-Pyridine ketone amino)-4-methyl-5-benzamide is an important organic chemicals, is widely used in the field of pesiticides, pharmaceuticals, materials and so on. In this paper, acetoacetanilide was used as raw material. a-Chloroacetoacetanilide was synthesized on the base of chlorinated reaction. I researched the process conditions about a-Chloroacetoacetanilide and thiourea solution as raw materials in the synthesis of 2-Amino-4-methyl-5-benzamide. Then I reasearched the process conditions about 2-Amino-4-methyl-5-benzamide and pyridine-4-carbonyl chloride as raw materials in the synthesis of 2-(N-Pyridine ketone amino)-4-methyl-5-benzamide.And I found the best optimum process conditions of the synthesis of a-Chloroacetoacetanilide,2-Amino-4-methyl-5-benzamide, 2-(N-Pyridine ketone amino)-4-methyl-5-benzamide.The optimun process conditions for preparing a-Chloroacetoacetanilide from acetoacetanilide and sulfurychloride were as follow:the molar ratio of a-Chloroacetoacetanilide to acetoacetanilide of 1:1.1, reaction temperature of 30℃, reaction time of 1.5h, the amount of dilute sulfuryl chloride of 12mL, dichloromethane of 40mL. Under this condition, the yield can reach 91.4%, and its purity can reach 98%.The optimun process conditions for preparing 2-Amino-4-methyl-5-benzamide from a-Chloroacetoacetanilide and thiourea solution was as follw:the molar ratio of a-Chloroacetoacetanilide to thiourea solution of 1:1.2, reaction time of 3h, reaction temperature of 90 ℃, concentated hydrochloric acid of 0.5mL, water of 35mL. Under this condition, the yield can reach 94%, and its purity can reach 99.2%.The optimun process conditions for preparing 2-(N-Pyridine ketone amino)-4-methyl-5-benzamide from 2-Amino-4-methyl-5-benzamide and pyridine-4-carbonyl chloride was as follow:the molar ratio of 2-Amino-4-methyl-5-benzamide to pyridine-4-carbonyl chloride of 1:1.3, triethylamine of 2mL, acetonitrile of 30mL, reaction temperature of 80, reaction time of 3.5h. Under this condition, the yield can reach 87.4%, its purity can reach 98.3%.
