Based on selection and optimization in the DNA extraction methods of edible oil at the same time, try to use qPCR, SSR, SNP,SCoT and other molecular technologies for carrying out the varieties traceability ofpeanut oil, so as to provide technical help for improving the guarantee system of thequalityofedible oil and maintaining the better market order ofedible oil.The main experiment results were as follows:To compare a large amount of the DNA extraction methods of the edible oil, including traditional CTAB, SDS, all kinds of n-hexane emulsification methods, Resin Oil DNA Extraction Kit, QiAamp FastDNA Stool Mini Kit, Resin DNA Extraction Kit was confirmed the best one to meet the requirements of the varieties traceability.Designing species specific primers of peanut according to internal standard gene sequences, optimize the qPCR detection system ofoil DNA.The result showed the correlation coefficient(R2) value between Cq value and differentdiluted concentrations of oil DNA reached up to 0.993, the qPCR standard curve could realize quantitative analysis of oil DNA.Using 9 kinds of main peanut varieties as the materials from Shandong, Henan andHebei province, the varieties traceability of edible oil was researched with the molecular marker of SSR, SNP, SCoT.The DNA templateofpeanut leaf was used for the preliminary screening of molecular marker technologies.The result showed that 4 of 20 pairs of SSR primers were polymorphic, including specific markers; 2 of 4 pairs of 2 kinds of SNP primers were polymorphic, the specific marker of LuHua 8 cultivar was the first kind,which matched with the AS-PCR reaction; all varieties tested coule be divided into two classes with the effect of the second kind; each of SCoT primers was polymorphic and could be used as specific markers for specific varieties. All the remaining primers through the preliminary screening were applied to all oil DNA templates and took advantage of the same polymorphiceffect,the result confirmed that the molecular markers couldbe used well to thevarieties traceability analysisof peanut oil.
