Due to their characteristic structure properties, chiral polycyclic skeletons are a kind of important core element and featured in a myriad of naturally occuring alkaloids and medicinally relevant compounds, which have exhibited infrequent biological activities and pharmacodynamics efficacy. Asymmetric cascade reactions have become one of vibrant research fields in organic synthesis because of their high efficiency and practical applicability, which avoided several expensive and time-consuming protection-deprotection processes, especially tedious intermediate purification steps. More and more researchers have paid their attention to the development of novel and effective cascade processes. Therefore, a series of elegant and pratical methods have been smoothly presented so far. We developed several newly-designed new asymmetric cascade reactions mediated by small molecular thiourea to achieve the efficient construction of various chiral polycyclic skeletons, such as chromanes and medium-sized lactones.
