Apricot kernel protein was hydrolyzed by pepsin, alkaline protease, neutral protease, trypsin and papain, respectively. The effect of apricot kernel protein enzymatic hydrolysates on proliferation of human breast cancer cell (MCF-7) was investigated. Four fractions with different molecular weight were prepared using ultrafiltration technology, and the effect of them on proliferation of MCF-7 cell was investgated. The amino acid composition and contents of four different fractions were analyzed. Meanwhile, the complexes of apricot kernel protein enzymatic hydrolysate and oleic acid were prepared, and the effect of the complexes on proliferation of MCF-7 cell was investgated.The main results were as follows:(1) The apricot kernel protein was hydrolyzed by pepsin, alkaline protease, neutral protease, trypsin and papain, respectively. The effect of apricot kernel protein enzymatic hydrolysates on proliferation of human breast cancer cell (MCF-7) was investigated. The results showed that enzymatic hydrolysates produced by alkaline protease, neutral protease, trypsin and papain exhibited inhibitory activity on proliferation of human breast cancer cell (MCF-7), and the papain hydrolysate showed the most significant effect, among them. As such, the papain hydrolysate was chosen for use in all subsequent experiments herein, and was called for apricot kernel protein enzymatic hydrolysate.(2) Apricot kernel protein enzymatic hydrolysates were separated by ultrafiltration into four fractions based on molecular weight,10KDa>MW>5KDa,5KDa>MW>3KDa,3KDa>MW>lKDa and MW<1KDa. The effects of the four fractions on MCF-7 cell proliferation were investgated. The results showed that the acticity of MW<1KDa fraction was the most significant, followed by 3KDa>MW>1KDa,5KDa>MW>3KDa and 10KDa>MW>5KDa fractions.(3) Amino acid composition and content analysis of the four fractions with different molecular weight showed that 18 kinds of amino acids were contained in all the four fractions, like tryptophan, histidine, glutamic acid etc.The content of hydrophilic amino acid in<1KDa fraction was the richest, followed by 1-3KDa, 3-5KDa,5-10KDa fractions. The content of hydrophobic amino acid in<1KDa fraction was also the highest. The results showed that the fractions with stronger activity, not only contained higher levels of content of hydrophilic amino acid, hydrophobic amino acids but also were more abundant. In addition lower molecular weight fractions had higher content of arginine.(4) The complex from apricot kernel protein enzymatic hydrolysate and oleic acid was prepared. The effect of the complex on MCF-7 cell proliferation was investgated. The results showed that the inhibitory effect of the apricot kernel protein enzymatic hydrolysate was enhanced after binding oleic acid.
