| With the increasing amount of plastic, the disadvantage of the traditional plastic is becoming more and more prominent, so white pollution has becoming a hot spot in the global scale. This study is mainly about thecyanoethyl modificated paper-targeting degradation microbes, the main contents and the results are as follows:(1) Use cellulose Congo red medium, after rich set training and times coated cloth, and underlined separation purification, filtered out 18 strains from different bacteria source which has cyanoethyl modificated paper degradation capacity, name them respectively number for A1, and A2, and A3, and A4, and A5, and A6, and B1, and B2, and B3, and B4, and B5, and B6, and C2, and C3, and C4, and C5, and C6, and C7.(2) Determination of cellulase activity of 18 samples, the results show that the best activities of sodium carboxymethyl cellulose enzymes is C2, A5, C5; filter paper enzyme activity is Al, B1, A4; β-glucosidase activity was C2, A5, A2. In its physiological and biochemical characteristics of these 7 strains detection and 16S rRNA sequence analysis, combined with colony morphology and physiological and biochemical characteristics, identification A1 samples belonging to Alcaligenes SP. A2 belongs to Ochrobactrum sp.A4 bacteria belonging to Brevibacillus sp.,A5 belonging to Pseudomonas Mendocina sp.,Bl bacteria to Pseudomonas sp.,C2 and C5 are belonging to Brevibacillus SP.(3)Degrading bacteria on degradation resulting from the content of reducing sugar and modified paper material degradation rate optimization of experimental conditions for the index. Of culture temperature and Tween-80 content, pH value, compound inoculation, incubation time, nitrogen sources and nitrogen content influence on the degradation of paper degradation ability and by orthogonal experiment to test the temperature, Tween-80 content, pH value and the shaking speed the degradation effects of size. Enzyme-producing results after tests confirmed that the highest combination of strains is C2 and A4, C5, and the proportion between them for the C2:A4:C5=1:2:1. Best medium:peptone 1g/L, Tween-800.3%, cyanide of ethyl-modified stock 10g,MgSO40.5g,K2HPO41g,NaC10.5g,pH=8,1000mL,12℃ distilled water sterilized 20min; best cultivation environment:temperature 33 ℃, shaking speed 140rpm, training time for 3d.Obtained a more excellent degrading bacterial mixed culture systems. And a preliminary investigation based on the basic features of the training system, biodegradation optimization improves cyanoethylation modified application of paper materials in environmental protection, and also fill this novel thermoplastic materials in the field of bio-degradation experiments gaps and lack of data. |
