Dendrobium officinale Kimura et Migo is extremely valuable resource, and yet it is still not being used efficiently nor properly. Its leafs and stem have similar compound.However, its leafs had often found unwanted. Therefore, this study mainly targets the Dendrobium officinale Kimura et Migo leaf’s specify chemical components and their pharmacodynamics in order to prepared Dendrobium officinale Kimura et Migo leaf Granules(DG). We have studied preparation process and quality control.Phenol-Sulphuric acid method was employed to detect the content of polysaccharides.A method based on pre-column derivatization HPLC to analyze monosaccharide contents in Polysaccharides Dendrobium officinale Kimura et Migo Leaf was establish. The polysaccharide were hydrolyzed into monosaccharides with hydrochloric acid(HCL). The monosaccharide derivatives obtained with 1-Phenyl-3-methyl-5-pyrazolone(PMP) were separated by HPLC using a gradient elution process. The monosaccharide compositions in Polysaccharides were composed of mannose, rhamnose,galacturonic acid, glucose, galactose, xylose and arabinose with a molar ratio of 4.31:0.24:0.76:1.00:0.83:0.16:0.72.This method is accurate, quick, has both good reproducibility and strong specificity, which can be used to determine the monosaccharides in polysaccharide of Dendrobium officinale Kimura et Migo Leaf.In this study, two extraction process was compared, On the base of single factor tests,the Box-Behnken Response Surface Analysis was adopted to optimize polysaccharides’ extraction technology technical parameter. Extraction rate was regard as response value and the dosage liquor radio, extraction time, and extraction frequency was done as factors.Result: The optimum extraction conditions were as fallows:extracted polysaccharides 2times with 25 times the amount of water for 70 s each time., and the extraction rate was88.52%. The adoption of Box-Behnken Response Surface Analysis is reasonable and effective to optimize the extraction technology of polysaccharides from Herba Dendrobii Officinalis Leaf, and the extraction rate coincides with the theoretical one.Comparison of both concentration method and drying method, Mixed the dry ointment power, dextrin and xylitol by the ratio of 2.92:2.92:0.96,with 90% ethanol as solvent, the granulars are easily formed. Pilot studies show DG process is stable,easy to operate,adapted to large-scale production.This subject studies on the quality standard of DG from traits, TLC and content determination and so on. Preliminary determination of standard content of DG for each bagful contains not less than 0.6g polysaccharide and not less than 0.2g Man. Accelerated test, illumination test and long-term test were used to study the stability of DG. The test results showed that the main indexes of DG had no obvious change after these texts.After this research, we have established methods to analyze monosaccharide contents in Polysaccharides Dendrobium officinale Kimura et Migo Leaf. which is accurate, quick,has both good reproducibility and strong specificity. The extraction process is much more effective and easy to operate compare to its traditional way. Three batchs of DG experiment is basically consistent with the experiment results, which can provide the basis for the enterprise production and the industrialization of DG.
