In this study, the chrysanthemum indicum oil is extracted from the chrysanthemum indicum of Changbai Mountain, using soxhlet extraction, ultrasound assisted extraction and supercritical CO2 extraction method, respectively. The optimum conditions and parameters were determined based on the single factor experiment, orthogonal test and Box-Behnken experimental design. Antioxidant activities of the chrysanthemum oil were investigated after the analysis of the chrysanthemum oil by GC-MS. The scavenging capacities to DPPH radicals, ABTS radicals, superoxide anion and hydroxyl radicals were measured, which provided theoretical basis of the applications in native antioxidant, supplements antioxidant and food addition agent. This study could lay the foundation for the further application of the chrysanthemum oil in food industries, pharmaceutical and Bath Items. The main results of the study were as follows:1. The oil from chrysanthemum indicum was extracted by the method of soxhelt with petroleum ether as solvent. The yield of chrysanthemum indicum oil was taken as index, which is effected by granularities, extraction temperature, extraction time and ratio of material to liquid. Based on single factor experiments, Box-Behnken experiment was designed among extraction temperature, extraction time and ratio of material. The results indicate: extraction temperature is 50℃, extraction time is 9.5h, and ratio of material is 1:40, the chrysanthemum indicum oil yield was 4.968%. To determine the component, the chemical components of chrysanthemum indicum oil was analyzed by GC-MS, a total of 43 compounds were detected in the chrysanthemum indicum oil, which accound for 78.52% of total volatile components, including alcohol, aldehyde, ester, alkane, alkene, ketone and others, the alcohol and alkene content are highest, 44.06% and 16.82%, respectively.2. The oil from chrysanthemum indicum was extracted by the method of ultrasound assisted extraction with petroleum ether as solvent and taking the yield of chrysanthemum indicum oil as index, the effects of granularities, extraction time, extraction temperature and solid to solvent ratio were investigated. Using an orthogonal array design based on single factor experiments. The results indicate: granularities is 60, extraction time is 15 min, extraction temperature is 25℃, and solid to solvent ratio is 1:20, the chrysanthemum indicum oil yield was 3.897%. To determine the component, the chemical components of chrysanthemum indicum oil was analyzed by GC-MS, a total of 45 compounds were detected in the chrysanthemum indicum oil, which accound for 60.11% of total volatile components, including alcohol, alkene, alkane, organic acid, ester, ketone, aldehyde and others, the alcohol and alkene content are highest, 28.62% and 7.22%, respectively.3. The oil from chrysanthemum indicum was extracted by the method of supercritical CO2 extraction method and taking the yield of chrysanthemum indicum oil as index, the effects of granularities, extraction time, extraction temperature and extraction pressure to chrysanthemum indicum oil yiled were investigated. Based on single factor experiments, Box-Behnken experiment was designed among the extraction time, extraction temperature and pressure. The results indicate that: extraction time is 150 min, extraction temperature is 45℃, and extraction pressure 30 MPa, chrysanthemum indicum oil more than 6.196%. To determine the component, the chemical components of chrysanthemum indicum oil was analyzed by GC-MS, a total of 53 compounds were detected in the chrysanthemum indicum oil, which accound for 89.46% of total volatile components, including alcohol, aldehyde, ester, alkane, alkene, ketone, organic acid, phenol and others, the alcohol, alkene and organic acid content are highest, 35.95%, 11.46%and 8.37%, respectively.In conclusion, the yield by the supercritical CO2 extraction method and the total volatile components are the highest among the three different methods. The method has advantages of less solvent pollution and easy process, which is the build basis to antioxidant activity study.4. Antioxidant activity of the chrysanthemum indicum oil was analyzed using supercritical CO2 extraction method. The results show that scavenging abilities of DPPH, ABTS radicals, superoxide anion and hydroxyl radicals were increasing along with the concentration of the chrysanthemum indicum oil increasing, and reducing power was increasing. When the concentration of chrysanthemum indicum oil was 2mg/m L, the superoxide anion scavenging was 98.38%.
