| Wild Actinidia arguta in Changbai Mountain,as a natural wild fruit resource of the same origin of medicine and food,is is not only rich in VC and practical cellulose,sugars,amino acids and other nutritional elements.Its fruit also contains certain functional ingredients such as flavonoids.A more in-depth study on it will not only benefit the development of wild fruit resources,but also provide theoretical support for the research and development of wild Actinidia arguta products in Changbai Mountains,which are both medicinal and edible,and have important scientific research value.At present,the extraction and development of total flavonoids from wild Actinidia arguta in Changbai Mountain is not perfect.And its in-depth research on its anti-oxidation effect has not been reported yet.Its scientific research and product development still have a lot of space.In this experiment,the total flavonoids of wild Actinidia arguta in Changbai Mountain were extracted and purified,and the extracts of different picking stages were identified by HPLC,and the antioxidant activity of flavonoids extracts from Actinidia chinensis at the most suitable picking stage was systematically studied.The following conclusions were reached:(1)According to the yield of total flavonoids,the optimum conditions of ultrasonic-microwave synergistic extraction with response surface were determined as follows:liquid/material ratio 25∶1(mL/g);ultrasonic power 350 W;ultrasonic time7 min;microwave power 360 W;microwave time 336 s.under the optimal conditions for 3 times test,the extraction yield of flavonoids could reach about(7.33±0.002)%.(2)Four kinds of macroporous adsorption resins were compared and their adsorption rates and resolution rates were analyzed under the same conditions.Finally,AB-8 resin was selected to purify wild Actinidia arguta.The sample solution containing 0.3150 mg/mL was loaded with 50 mL wet method and adsorbed at the flow rate of 1.0 mL/min.After the sample was finished,the impurity was washed with70mL water and then eluted with 70%ethanol of 70mL with 2.0 mL/min.Under the optimum purification conditions,the purity of total flavonoids was 81.43%.(3)According to the optimum extraction technology and purification conditions,the content and purity of total flavonoids in wild Actinidia arguta fruits at different picking periods were compared and analyzed,and the optimum experimental materials were selected to further study the antioxidant activity.(4)The main components of the extracts of Wild Actinidia arguta in Changbai Mountain in different picking periods were identified and analyzed by HPLC.Rutin,quercetin and isorhamnetin were included in the total flavonoids of wild Actinidia arguta.(5)The antioxidant activity of total flavonoids of Actinidia arguta before and after purification was studied,and the same VC was tested respectively.The purified Actinidia arguta flavonoids on DPPH free radicals IC50 0.024 mg/mL,VC IC50 0.049,indicating that the removal rate of flavonoids from Actinidia arguta was higher than that of VC in the removal effect of DPPH free radicals.Through the comparison and analysis of the clearance rate curve,it can be found that the scavenging ability of the purified elution solution to OH free radicals is better than that of VC solution.After purification,the IC50 of Actinidia arguta flavonoids on O2-free radicals was 0.039mg/mL,the IC50 of VC was 0.021 mg/mL,and the removal ability was worse than that of Vc,but it was always higher than the scavenging ability of crude extract to O2-free radicals before purification.It can be purified that the removal of flavonoids from the Actinidia arguta has improved the scavenging effect of DPPH,OH and O2-free radicals,and a good alternative to VC. |
