Multisubstituted pyran pyrazoles, sprio [indoline-1,2,4-triazolo[1,5-a] pyrimidine] and thiazole hydrazone compounds are widely exist in nature, of which the similar skeleton structure was found in many drugs and natural products with biological activities. Studies on bioactivities showed that these compounds have good analgesia, antibacterial, antitumor, anti-inflammatory, deinsectization and other biological activities. Its wide range of biological activities and medicinal value were used in medicine and pesticide fields. Multi-component reactions(MCRs) constitute a versatile class of organic chemistry widely employed for the synthesis of complex heterocycles and natural products. This method of reaction have attracted considerable attention because of their valuable features such as atom economy, straightforward reaction design, energy savings, and reduced waste. So it is significant to find out a simple, efficient, good yield, characteristics of atomic economy method for the systhesis of these compouds. This paper will be divided into the following two parts.The first part is a review. Introduces the development status of multisubstituted pyran pyrazoles, spiro [indoline-1,2,4-triazolo[1,5-a]pyrimidine] and thiazole-hydrazone compounds, the research purpose, meaning, as well as the research content and plan of this topic.The second part is about experiment, it introuduced the major work. 1. Facial one-pot, three-component synthesis of thiazole compounds by the reactions of aldehyde/ketone, thiosemicarbazide and chlorinated carboxylic ester derivativesA series of thiazolyl-hydrazone derivatives were synthesized via three-component one-pot reaction of aldehyde/ketone, thiosemicarbazide and chlorinated β-keto ester in EtOH, using CH3 COONa as a base catalyst. 2. Highly efficient one-pot synthesis of multisubstituted bispyrano[2,3-c]pyrazoleA series of multisubstituted bispyrano[2,3-c]pyrazole derivatives were synthesized via four-component one-pot reaction of Methyl acetoacetate, hydrazine derivatives, N-Methyl-1-(methylthio)-2-nitroethylen-1-amine and p-phthalaldehyde/m-phthalaldehyde in ethanol under refluxing condition catalyzed by Et3 N. 3. Cs2CO3-promoted one-pot synthesis of sprio-[indoline-1,2,4-triazolo[1,5-a] Pyrimidine] and sprio-[indoline-benzimidazole[1,2-a]pyrimidine] compounds.The sprio-[indoline-1,2,4-triazolo[1,5-a]pyrimidine] and sprio-[indoline-benzimidazole[1,2-a] pyrimidine] compounds were synthesized by three-component one-pot reaction of 3-amino-1,2,4-triazole/2-aminobenzimidazole, isatin, and malononitrile in the presence of Cs2CO3(20 mol %) as a catalyst.
