Taxol possess a prominent curative effect to many different cancer, so it has been much attention. At present, the main source of taxol is extracted from the yew tree, but because of the scarcity of natural resources yew and the contain of taxol in yew tree were very low, which is far from to satisfy market needs. Using yew endophytic fungi fermentation in recent years is a new way to procure taxol。optimized fermentation conditions of high-yield strain of taxol is one of the effective ways of improve the endophyte taxol biosynthesis of.The test take strains HDF35-8-6 as the tested strains , to study and research carbon, nitrogen in fermentation medium and fermentation process parameters and fed-batch fermentation, to enhance the amount of taxol of taxol-producing strains biosynthesis, early realization of endophytic fungi fermentation of industrial production of taxol. Test results show that:1、Adopt Plackett-Burman design have selected significant influencing factor of strains HDF35-8-6 taxol biosynthesis from carbon source, nitrogen source and the fermentation process parameters.Carbon source as follows:glucose, sucrose and maltose; nitrogen as follows:ammonium acetate,ammonium chloride and soybean meal hydroly -zeate; fermentation parameters:shaker speed, temperature and initial fermentation medium pH;2、Useing the response surface analysis to evaluate the main impact factors and optimization has been the impact of strain HDF35-8-6 Production of Taxol quadratic polynomial regression model. SAS9.1 software using the model test of significance, test results showed that when glucose 6.76g/L,sucrose 9.48g/L,maltose 6.89g/L, ammonium acetate 0.87g/L, ammonium chloride, 0.52g/L, soybean meal hydrolyzate 18.46mL/L, shaking speed 119r/min,fermentation temperature 28.8℃, the initial fermentation medium pH6.7, the strain HDF35-8-6 taxol biosynthesis has reached the maximum value for the 539.74±4.63μg/L, than optimizing the pre-strain HDF35-8-6 synthesis of paclitaxel was enhanced by 20.58%;3、To determine the strain HDF35-8-6 production of Taxol fermentation batch process, that is the beginning of fermentation (No. 0d) by adding a one-time glucose 6.84g/L, sucrose 7.45g/L, maltose 5.43g/L, ammonium acetate 0.79g/L, ammonium chloride, 0.46g/L, soybean meal hydrolyzate 16.62mL/L; after fermentation 8d cultured S-7 to the liquid fermentation medium sugar added 3.48g/L, maltose 2.52g/L, ammonium acetate 0.125g/L, ammonium chloride, 0.074g/L, soybean meal hydrolyzate 2.66mL/L. The fed-batch process experiment results show that taxol production has been greatly improved by the previous batch of 539.74±4.63μg/L increased to 613.70±3.64μg/L, increased14.1℅; than before optimization strains HDF35-8-6 (447.42μg /L) increased the taxol yield 37.14 percent.
