With the development of Chinese shipping industry,the problem of the harm of bacteria in ships’ballast water has become more and more serious. Traditional methods about Bacteria detection in ballast water mainly include flow cytometry, fluorescence microscopy and biochemical methods, but there are some disadvantages in these methods such as too large in size, too complex to operation,time-consuming, unable to achieve real-time detection and so on.In view of this, A whole PDMS microfluidic chip bacteria rapid detection method that the content of bacteria is be determined by dectect the ATP with microfluidic chip as the bacteria detection platform is put forward in this paper. In this method, with microfluidic chip as the bacteria detection platform, characterized by ATP content in the detection of bacteria bacteria. A rapid detection of bacteria in ballast water system based on microfluidic chip is successfully designed and the accuracy of the system is verified by a large number of experiments in this paper. The main work of this paper includes:(1) Through the analysis of principle, the design and construction of detection system are completed. Detection system consists of hardware and software. Hardware components include low light detection, microfluidic chips, systems and mechanical structures; software programming is completed through LabVIEW graphical programming software, it includes conversion, data acquisition, processing software pulse counting and light intensity display.(2) The performance’s tests of detection system is completed including detection system the end of test, test different types of Microfluidic Chip and standard ATP detection test solution. Detection system test at the end of test mainly test system’s background noise; Microfluidic Chip testing is to test chip fluorescent effect on the test results; the standard ATP solutions testing is done mainly for system accuracy and sensitivity analysis. Test results showed that the detection system can quickly and accurately detect the concentration of bacteria.(3) The different bacteria detection and analysis of samples are completed in this paper. Detection and analysis of solution of different concentration of Escherichia coli can be achieved by combining concentrated approach, and the system can effectively detect the original concentration of 200cfu/100mL which meets the D-2 standard. At the same time, the effects on detection in the presence of algae are also studied in this paper.
