Raman spectrum is molecular scattering spectrum with advantages of non-destructive, rich information, no sample preparation etc. Using Raman spectroscopic analysis combined with confocal microscopy, not only the Raman spectrum signals of different micro area from samples in the same plane, but also in different depth can be observed, and the measured sample can be tomography reaching an "optical sectioning" effect without damaging the sample. Confocal Raman spectroscopy has become an important means of observation in many research fields such as food, chemical, nano materials, biological, forensic evidence, etc. Moreover, in testing of inclusions of mineral or gemstone, especially calligraphy and painting and the coating layer of some objects, it is also a very effective tool.The main research is on the measurement and control technology of laser confocal Raman spectroscopy based on the principle of confocal focusing. A measurement and control system is designed, and in the system the host computer is used to coordinate the various components of laser confocal microscope and Raman detection system, the image information of microscopic and Raman will be collected and processed, realizing three-dimensional map imaging of the micro area sample with high precision.The main research works in the paper are:1. The software structure of the confocal Raman spectroscopy is designed by leaning and analyzing the software of spectroscopy existing in domestic and foreign. While the system software and interface is optimized in Microsoft Visual Studio 2010 software development environment using multiple documents mode according to object-oriented design requirements.2. The scanning control software of confocal Raman spectroscopy is designed. According to the principle features of confocal Raman spectroscopy, a set of the scanning control software is projected to achieve hardware initialization, parameter setting, data acquisition and processing, displaying and file saving. Take advantage of the software trigger mode to achieve focusing and acquainting comprehensive information with microscopic and spectral. In addition capabilities of single-point spectrum mining and spectral imaging based of peak intensity are realized.3. It is verified that the software is practical and stability using confocal Raman spectroscopy experimental apparatus built in the laboratory to scan image of the sample under the control of the software.
