Chinese Silky chicken is a very precious indigenous chicken breed of China. It is famous because of its outstanding nutritional and medical value,which was proved to be related with the existing of aboundant melanin. Both white plumage color Silky and black plumage color Silky chicken have melanin secreting and deposition in the skin,muscle and bone (black skin,black muscle and black bone).However,the melanin doesn’t extend to the feather in Taihe chicken. The study of genetic basis underline will be important to the discovering the molecular mechanism of melanin deposition.We assume that the loci controlling the relative difference of white and black feather are all homozygosis in both chicken breeds. Therefore,the internal cross of the F1 individuals generated by the crossing of White Silky and Black Silky will induce the segregation of relative traits.According to this supporse,the following experiments were conducted:1.Three Black Jinshui Silky cocks(Black(?)) intercrossed(×) with three White Jinshui Silky hens (White♀);Three Black Jinshui Hens(Black♀) intercrossed with three White Jinshui Silky cocks (White(?)).This cross experiment yielded 29 F1 individuals, with two different types of feather color, black or piebald.2. Four intercross experiments:Blackc(?)×Black♀,Piebald(?)×Piebald♀, Black(?)×Piebald♀,Piebald(?)×Black♀were carried out in F1 progeny from Black(?)×White♀;And F1 Black(?) progeny from White(?)×Black♀were intercrossed with Black♀and Piebald♀progeny from Black(?)×White♀.This cross experiment yielded 121 F2 individuals with trait segeration,the number of all the population about 147.3.Genotype distribution and allele frequency at polymorphic sites of Mclr gene and EDNRB2 gene in different chicken traits.linkage analysis SNPs of EDNRB2 gene and the feather color using CRIMAP software. Constructed EDNRB2 gene haplotype block; and the relationship test between SNPs of EDNRB2 gene and feather color using pearson chi-square.4.Recessive white feather detection method established by the PCR,in the fourth intron of TYR gene have retroviral insertional mutagenesis with birds lend to Recessive white feather, experimental results show that parents of black feathers are no retroviral insertion, but All white feather have retroviral insertionThe present results revealed that other additional loci were involved in the controlling of the relative difference of White and Black feather besides of the C locus. Therefore,the present results established a basis for the next study.
