Hydrangea[Hydrangea macrophylla (Thunb.)Ser] has large inflorescence, rich and ruby color. It has long flowering period between 1-2 months with adequate nutrients. Hydrangea is a potted flower and good for landscaping. In the United States, about 32 million U.S. dollars in sales annually (Jones KD,2006), and plants for sale are higher than 30 U.S. dollars/basin. China has introduced several cultivars of H. macrophylla in recent years. The traditional Hydrangea cuttings use for spring and autumn cannot meet the needs of the annual demand. Its propagation cycle is long thus difficult to meet demands.and compete for the market.We introduced new excellent hydrangea cultivars as materials for the tissue culture and use of stem explants for rapid propagation of axillary bud. This greatly improved the survival rate. Appropriate technology is required to generate and reshape the mini potted hydrangea plant. To do this first we explored Wuhan to identify a potted hydrangea retardant. The autumn leaf spot pathogen was identified as the plant most serious retardant.Stems with axillaries bud of Hydrangea macrophylla were used as explants to conduce in vitro culture. Different hormone combination treatment at different concentrations of’Hdy5’(code-named cultivar of Hydrangea macrophylla)were used. Experimental results show that the average coefficient of the highest proliferation medium for B5+6-BA1.5 mg/L+GA30.6mg/L, pH5.5-6.0, agar 7.5 g/L, glucose 30 g/L, is 3.67 at induction rate of 100%. The most appropriate proliferation medium for ’Hyd5’was also widely used in several other cultivars. Overall,’Hydl’,’Hyd4’,’Hdy5’,’ Hyd8’growth robustly,’Hyd3’and’Hyd6’shoots a lot but yellowish,’Hyd9’relatively thin. Therefore, the best way to optimize proliferation for’Hdy3’,’Hdy6’,’Hyd9’remains to be continued.Tissue culture, seedling and transplanting:The best culture medium for the proliferation of transplanted seedlings is:perlite 2cm with sugar 5g/L of 1/2B5 liquid medium and inoculation density of 8/bottle. Although the economical rooting media: perlite 2cm+water medium(2cm) had a survival rate higher 90%, and cost is only 0.01 yuan/bottle; the most suitable matrix for transplanting is the perlite:Northeast peat were (1:3) copies; but the medium:perlite+liquid medium(2cm) can quickly obtain a robust growth of tissue culture, as the media cost only a fraction of the total cost of production, so more suitable for large scale production of high quality seedlings. With proliferation, transplanted seedlings were 3cm higher than commodity seedlings and transplanting survival rate was 90%.Potted hydrangea dwarf cultivation:The dwarf potted Hydrangea agent was diluted 300-fold 50% of the CCC liquid according to the protocols of light agricultural chemical Co., Ltd. Sichuan, China production.. Photoperiod treatment of the plant was done when 83.2% of the control reached average height of 6.0cm, bud when the plant height is 15.07cm, height was only 75.5%. In March the budding rate was 80% higher than in the control 70%. The average height was about 18cm (inflorescence plus height) when flowering occurred at May 10th.Greenhouse hydrangeas Disease Identification:Leaf spot in autumn severely affected the Greenhouse potted hydrangea. The pathogen was isolated, and the pathogen which caused the leaf spot was found in two, one with the Gen bank of Phoma putaminum, Phoma multirostrata matched of 100%, were imperfect fungi Amon stem Sphaeropsidales point spp. Another in Genbank matched Cercospora fukushiana, Cercospora penzigii, Cercospora kikuchii of 100%, are semi-known fungus Alternaria Gang Portacaval Sphaeropsidales Cercospora spp. Alternate useing of a variety of fungicides during cultivation, application of concentration can be in accordance with its instructions; when a large number of water spray as far as possible in advance to ensure that the evening has no leaves and soil surface moisture in summer, so as to avoid creation of a good breeding and propagation environment for virus; diseased leaves should be promptly removed, moderate watering, and increase nitrogen fertilizer to promote leaf update.
