Ten kinds of Dendrobium officinale Kimura et Migo were planted in Quanmei Bioscience Inc to study their agronomic characters, yields, polysaccharide contents and cellulose contents, and then filtrate the high yield and high quality Dendrobium officinale Kimura et Migo—DEDR0203. We explored the difference of physiological indexes in Dendrobium officinale Kimura et Migo, analyzed the relation among yields, polysaccharide contents, cellulose contents and physiological indexes. Next, tissue culture and rapid propagation system and integrated cultivation technique of Dendrobium officinale Kimura et Migo were set up.. The main results are as follows.Firstly, we evaluated the ten kinds of Dendrobium officinale Kimura et Migo systematically. According to evaluating results, we filtrated Dendrobium officinale Kimura et Migo introduced from Yunan numbered as DEDR0203, whose polysacch-aride content achieved 268.69μg/mg and polysaccharidea ccumulation achieved 552.40 kg/ha, both of them were the highest.Secendly, when Dendrobium officinale Kimura et Migo were planted, results showed that as follows. Growth and quality of Dendrobium officinale Kimura et Migo were proportional to their chlorophyll. Metabolic capability and resistance ability of ageing of Dendrobium officinale Kimura et Migo were enhancing to promote their polysaccharide accumulation with contents of soluble protein being higher and higher. Carbohydrate metabolism of Dendrobium officinale Kimura et Migo were enhancing to promote their polysaccharide accumulation with contents of soluble sugar being higher and higher, on the other hand, environmental adaptability of Dendrobium officinale Kimura et Migo were proportional to their contents of soluble sugar, reducing the negative effect on the growth environment adaptability. Dued to can not adapt to the new environment, contents of proline were enhancing, which inhibited the growth and polysaccharide synthesis, even qualities of Dendrobium officinale Kimura et Migo were affected. Environmental adaptability were promoting with activity of CAT and SOD strengthening, which promote the growth and development of Dendrobium officinale Kimura et Migo, in the end promote their yields and polysaccharide accumulation.Thirdly, methods were studied to improving germination rate, appreciation rate and rooting rate of Dendrobium officinale Kimura et Migo, and the results showed that explants were inoculated in the medium as MS+NAA 0.2 mg/L+IBA 1.0 mg/L +potato 100 g/L,25-27℃,1500-2000 Lux,12 hours per day whose inductivity achieved 90%. Propagation material were inoculated in the medium as MS+KT 0.2 mg/L+NAA 0.1 mg/L with potato 100 g/L in 35-40 days, whose proliferation multiple achieved 4.8. Tissue culture seedling were inoculated in the medium as MS+ NAA 0.5 mg/L+IBA 2 mg/L with banana 70 g/L and potato 100 g/L and AC 100 mg/L in 50 days, whose rooting rate achieved 100%.Finally, we studied factors to survival rate, growth and development of Dendrobium officinale Kimura et Migo, the results showed as follows. Before tissue culture seedling being planted, domesticated in the bottles in 10 days and then domesticated in the open air in 5 days, in consequence, the rooting rates were the highest. When the ratio of aqua-plant and moss was 3 to 1, the survival rates, growthes vigour and yields of Dendrobium officinale Kimura et Migo were the highest. Dendrobium officinale Kimura et Migo seedlings in shading rate of 40% developed more rapidly than seedlings in other shading rates. If Dendrobium officinale Kimura et Migo seedlings were planted during May to June or October to November, whose survival rates were the highest, after two years, Dendrobium officinale Kimura et Migo would be harvested.
