Euphorbia lathyris L. (Euphorbiaceae), is biennial herbage which originates Europe, mostly artificial cultivated. It distributes in all parts of china and adapt to sunny slopes. The dry seeds are the medicinal parts. It can eliminate the brake blood and estate water swelling. It can be used to cure various diseases including hydropsy, abdominal distention, phlegm-rheum, accumulations and gatherings, blood stasis, menstrual block, fecal and urinary stoppage, stubborn lichen, wart, sores and swollen welling-abscesses, snake bite, hepatocirrhosis, and leukemia, etc. In recent years, schistosomiasis reappears. The fruit of Euphorbia lathyris L. is the effective to treat patients with schistosomiasis in late course. It can also suppress tumor.Therefore, the demand for the seeds to culture Euphorbia lathyris L. on market is increasing rapidly accompanied by skyrocketing of expense for its fruit. A rapid growth in demand for the seeds, the price is increasing year by year.Euphorbia lathyris L. is a ideal energy plant.The oil content of its seeds reaches as high as 43-48%. The main composition of the seeds is fatty acid, reached 99%, including oleic acid and hexadecanoic acid, linoleic acid, etc. C16 and C18 account for most. Oleic acid content is 83%. It contains a lot of fermentable sugarsthat are fermented to produce alcohol. The residue is used as feed. In addition to high oil content of its seeds, its plants also have high oil-containing rate of 9.25. The whole plant can be used for petroleum refining. The skill of refining petroleum is very proficient at home and abroad. Therefore, Euphorbia lathyris L. has a broad development prospects and important economic value as an ideal energy plants.Euphorbia lathyris L. breeds by the seeds, in the wild. The production of herbs is reduced with breeding by seeds. In addition, in the seedling stage, likubin easily occurs while calico and tan disease happen in the late-mid period. Those diseases cause the reduction of production. Therefore, It is imperative to develop effective strategies against these diseases harmful to Euphorbia lathyris L through biotechnology. to solve the above problem by biotech as soon as possible.The research, based on Euphorbia lathyris L from Yuzhou, Henan province, takes MS as the basic medium (0.7% AGAR,1.5% sucrose), adopting the method of planting tissue. The rapid propagation path was set up by using different concentrations of 6-BA, while regeneration system was established by the combination of 6-BA and NAA of different concentrations. One advantage of this study is reduction of amount of seeds used for cultivation. This indirectly increases its production.The study also builds a solid foundation for development of genetic transformation system and transgenic breeding of this plant in the future. The main research results were as follows:1. Seeds sterilization and sterile seedling preparation:The peeled seeds were cultivated in 1/2MS, were immersed in 75% of ethanol for 1 min and disinfected in 0.1% HgCl2 for 6min prior to cultivation in 1/2MS. The temperature was controlled at (26±1℃), with light intensity of 15-20001x, and illumination time for 16h/d. The bacteria rate was lower than 0.55%. The sterile seedlings of 15d were cut into small sections to be explants.2. Establishment of rapid reproduction system:Stem tip, stem the upper and stem bottom were used to select the optimal explants. The research showed that meristem tip and stem upside were grown well and are therefore selected as the best explants for the further studies.The rapid propagation buds were induced in MS+0.3mg/L6-BA. The budding rate reached up to 53%. These buds with fast budding rate were cultivated in MS+0.2-0.4mg/L6-BA.1/2MS medium make the buds strong. Good buds were cultivated in 1/2MS+0.2 mg/LNAA to root. The rooting rate was 80% after 60d. The root amount was 12.2 every seeding. The selected plants were transplanted in greenhouse. Finally the good plants were cultivated in the field.3. Establishment of regeneration system:Stem tip, the upper stem and stem bottom were used to select the optimal explants. The research showed that stem tip, stem upside were the best candidates.The Callus was induced in MS+1.0mg/L6-BA+0.2mg/LNAA. The induction rate reached 36.8%. The callus was propagated in the same medium. The average number of shoots was 3.33 every explant. Good buds were cultivated in 1/2MS+0.2 mg/LNAA to root. The rooting rate was 80% after 60d. The root amount was 12.2 every seeding. The selected plants are transplanted in greenhouse. Finally the good plants are cultivated to the field.
