Rose?Rosa chinensis?:is one of the representative plants of Rosaceae family Rosa genus.Rose is perennial deciduous or evergreen shrub,which is one of the most popular ornamental flowers.Because of the variety of color,fragrance,pattern and long period of flowering?blooming throughout the year when the external conditions are appropriate?,it is one of the most important cut flowers in the world with high commercial ornamental value and scientific research value.In this experiment,the rapid propagation system of two modern popular rose varieties was established,due to which,virus-free seedlings were obtained for other 3 popular rose cultivars.Then leaf veins and leaves were used as explants to improve the system of embryogenesis and regeneration of rose somatic embryos.It provides more theoretical basis for the faster and better construction of rose fast propagation system and regeneration system.The results of this experiment are as follows:1?Construction of rapid breeding system in roseIn this experiment,modern rose varieties"Little Rose"and"Blue Storm"were selected as experimental materials.The propagation speed and growth state of axillary buds were not only determined by the genotype of the materials tested,but also affected by the position where explants come from.In order to screen the optimal medium,the plant tissue culture was carried out by using the shoot segments with buds from the branches of small rose and blue storm as explants.The experimental results showed that:rapid proliferation medium(MS+1.0mg/L 6-BA+0.01mg/L NAA+0.1mg/L GA3+30 g/L Sucrose+7.5g/L Agar+1mg/L ZPL)was the best medium for proliferation and seedling growth.2?Construction of embryogenesis and regeneration system in roseFirst,we tried to establish embryogenesis and regeneration system with leaves and petioles from two modern cultivar“Small Rose”and“Lavender Wreath”.The experimental results showed that the genotype and explants type,culture light condition,the type and concentration ratio of hormones in the medium had important influence on the induction of somatic embryos and the initiation of intact regenerated plants.Under dark culture conditions,petioles of“Little Rose”and“Lavender Wreath”were induced on the medium?MS+5.0mg/L 2,4-D+10mg/L Ag NO3+400mg/L L-p+30g/L Glucose+3.0g/L GEL+1mg/L ZPL?,Callus induction rates were as high as 81.6%and 74.25%,respectively.Two weeks later,they were moved to the medium?MS+0.6mg/L 6-BA+0.02mg/L NAA+30g/L Glucose+3.0g/L GEL?.The callus grew green and strong after 12 weeks,but no somatic embryos were produced.Then the explants were made with veins from four cultivars-“Little Rose”?“Lavender wreath”?“Purple Spirit”?“The Blue of the Earth”.Under dark conditions in the T1 medium for one week,and under light for two weeks,and then moved to the medium?MS+1.0mg/L TDZ+0.01mg/L NAA+0.1mg/L GA3+30g/L Glucose+3.0g/L GEL?under light.Four weeks later,only the callus of"Blue of the Earth"produced embryogenic callus with an induction rate of 29.20%.Embryonic callus and somatic embryos produced by"Blue of the Earth"can sprout rapidly in the medium?1/2MS+1.0mg/L 6-BA+0.01mg/L NAA+30g/L Glucose+3.0g/L GEL?with a budding rate of 100%,After 4 weeks germination into seedlings,buds were transplanted into the B1 medium under light culture.After growing to 4-5 cm high,seedlings were transferred to rooting S2 medium.The rooting rate was up to 100%.
