Pleurotus cornucopiae is one of the important commercial varieties of mushroom andthe color trait is important commodities trait. researches shown that its color traits wasquantitative trait controlled by multiple gene. Blue brown strain of Pleurotus cornucopiaeCCMSSC00406saved at China Center for Mushroom Spawn Standards and Control (CCMSSC)was parent.34monokaryons were obtained from spore prints by serial dilution of basidiosporesin distilled water. F1generation was constructed by self-crossing between monokaryons and get125dikaryons. The color of F1generations was measured using Spectrophoto Meter(CM-2600D) and measurement data was analyzed for further. Combined with measured dataand Observation records strains of dark brown were screened from F1generation group. In thesame way the second self-crossing generation was constructed. Fruit-body color of F2wasvisually observed and recorded for analysis. Finally28dark brown strains were acquired.The42strains selected from the group were performed SSR validate genetic diversityanalysis and antagonism. The results shown that406-133and406-99、406-133-8and406-99-8、406-77-12and406-77-6、406-133-2and406-99-12didn’t appear antagonistic phenomenon,andthe genetic similarity betwen each other were very high and were in the same groups in theclustering analysis. So they coud be considered were the same strain for each other.In order to screen the resistant strains, the35tested strains were made a comprehensiveanalysis of laboratory resistance. Taking mushroom mycelium confrontation with pathogens, thedegree of antagonism is used for evaluation of resistance, and the statistical resistance level wasdefined for the evaluation of resistance. The resistance levels of35strains were significantlydifferent. The dark strains resistance were higher than the light strains.
