Animal growth is a complex biological metabolic process, which is affected byvarious factors such as genotype, hormones, nutrition, environment, ect. The effectsof these factors on animal growth are carried out by influencing animal endocrinesystem directly or indirectly. Studies have showed that plant flavonoids and saponinscan regulate the growth of animals. Therefore, further screening of natural activeingredients involved in animal central regulation is of practical significance.Acanthopanax contains a variety of saponins, polysaccharides, flavonoids and otherpharmacological composition, and have the function of gonadal, fatigue resistanceand preventing memory deficits. In addition, it can also can improve the IFN levels inthe body, and strengthen the body immunity.Previous research mainly focused on summing up feeding experiments, whichextends test period and restricts understanding of regulation mechanism. Screeningnatural active ingredients involved animal central regulation by in vitro blood-brainbarrier model is a faster shortcut method. This experiment focused on studying newtechnology of pig growth regulation, to determine the permeation rate ofacanthopanax glycosides B and E by in vitro co-culture BBB model.The ternary landrace was selected to isolate and culture brain microvascularendothelial cells and astrocytes by twice enzyme digestion, BSA and Percolldiscontinuous gradient centrifugation, and then co-culture these two kinds of cells toestablish in vitro blood brain barrier model. Using4h leakage test, rutin membranepermeability test and resistance of across the cell membrane (TEER) measurement tovalue its quality. Finally, the model was used to determine the permeation rate ofacanthopanax glycosides B and E after the quality being valued.The results showed that cells moved away from the brain capillaries after24hculturing, the cell morphological observation found that cells showed short spindle,colony was a typical "pebbles" and regional monolayer growing.6-7days later, cellsjoined together. Ⅷ factor antigen showed positive. Astrocytes swelled obviously,mainly were a polygon or fusiform, GFAP-FITC immunofluorescence staining waspositive.co-culture model showed higher across the cell membrane resistance and lowpermeability, and acanthopanax glycosides B can efficiently pass the blood brain barrier, and the permeability was the best under the concentration of50mg/L.Conclusion was determined as follows: this experiment illustrates the success inbuilding the high quality of in vitro model of pig blood brain barrier, and we got thatacanthopanax glycosides B can pass the blood brain barrier, which provide good prooffor following study on its mechanism and screening of other natural active ingredientrefer to growth.
