In this study, Da-tong Chinese onion-alliums medium for testing of differentmaterials, factor in selecting explants, hormone type, carbon source, cultureconditions from unfertilized impact on culture, and the results showed that:1、Most pollen mother cell of bud, which is in central of involucre,when involucresof the Da-tong Chinese onion-alliums is cracking, the bud is being open at the top, isin the late mononuclear stage, the mother cell of bud ovary in central is more In tetradstage, callus induction of the ovary with inoculation is the best in this period.2、In the screening test of basic culture medium, the three culture medium of MS, B5,N6can induce ovary to produce callus, the callus have differences form differentculture medium, culture medium of MS is best, culture medium of B5is followed,culture medium of N6is no callus.3、 Da-tong Chinese onion-alliums was treated by low temperature range0to7dUnder4℃, the cryogenic treatment has certain influence on the ovary callusformation, which the low temperature treatment with3d is best; the dark treatmentwith35℃is favorable for the formation of callus for ovary inoculation, heat stroketreatment with5d is best in induction rate. Generally high temperature heat stroketreatment is better than the low temperature pretreatment.4、The effect of unfertilized chicken onion cultivation sucrose than maltose in theapplication, the callus induction rate is the highest with75mg/L sucrose. the budinduction rate is best when sucrose concentration of50mg/L.5、The regeneration system was set up for onion-alliums explants with unfertilizedovary. The callus differentiation medium is that MS+2mg/L2,4-D+3mg/L6-BA.Differentiation medium is that MS+1mg/L1TDZ. Rooting medium is that MS+0.5mg/LNAA.
