Soybean (Glycine max) is one of the worldwide planted oil crops. It plays an important rolein the agricultural industry structure and people’s lives. But its production seriously affectted bya variety of environmental stresses, especially drought stress. Therefore, exploring the ability fordrought resistant of soybean and cultivating new strains to improve production is so important.The9-cis-epoxy-carotenoid dioxygenases (NCEDs) catalyse the rate-limiting step in ABAbiosynthesis. The expression of NCED gene not only starts the ABA accumulate and decisionsthe capacity of ABA accumulation. ABA signaling affects drought tolerance of soybean. In thisstudy, I transformed the vector of the GmNCED1gene into Arabidopsis and soybean. It canincreases strong tolerance to drought and high-salt. In addition, the study transformed the vectorof GmNCED1promoter and GUS gene fusion into tobacco. By analyzing the changes of GUSexpression activity in transgenic tobacco under a variety of abiotic stresses, verified theGmNCED1expression patterns. The results of this study are as follows:1. Ten transgene lines we get from overexpression transgenic Arbidopsis thaliana and twotransgene lines from transgenic Arbidopsis thaliana mutant that transformed by florial dip.2. The T3overexpression and gene-covering transgenic lines were testing by germinationrat and growth state when they are in different stress treatment, such as, ABA, PEG, salt. Theresults showed that GmNCED1enhanced transgenic plant resistance to ABA and PEG mightily.3. Overexpression of Arabidopsis growing better than the wild-type and gene-covering ofArabidopsis growing better than mutant lines after dealing with drought stress and salt stress.The soluble sugar content analysis showed that GmNCED1gene reduced the soluble sugarcontent. The chlorophyll content analysis showed that GmNCED1reduced the decomposition ofchlorophyll. Further illustrated GmNCED1gene play a role on resistance drought and salt., thusimproving plants tolerance on stress.4. Transformed the GmNCED1gene into soybean. I obtained seven T0and three T1transgenic lines. By southern blot analysis method, I recived one T2transgenic soybean.5. The transgenic tobacco had been stained by GUS. The result showed that: GmNCED1gene promoter regulated the expression of GUS under PEG, ABA, salt and saline stress. And theexpresstion is highest in leaves. I analysised the quantitative of GUS, compared to no treatment,the expression level of GUS respectively induced by ABA and PEG is2.8times and2.5times.
