Chionanthus retusus Lindl.et Paxt. which is also known as radish flowers, isOleaceae, tassel tree genus and deciduous tree. It is the second national protectedplants, mainly distributed in China’s Gansu, Shaanxi, Shanxi, Hebei, south of Henan toYunnan, Sichuan, Guangdong, Fujian, Taiwan etc. Tassel tree flowers contain manyflavonoids and three terpenoids. It can be on behalf of the tea, and have somemedicinal value. The material is good. It can be made as appliances and can be usedfor joinery; the cpntent of oil of the fruit is high. It is an important energy plants.Chionanthus retusus Lindl.et Paxt. have luxuriant foliage and spreading branches. Theflower is big and beautiful. It is an excellent afforestation and ornamamental trees. Thebreeding cycle of the traditional breeding methods of Chionanthus retusus Lindl.etPaxt. is long and the coefficient is low. Up to now, the culture of organizationChionanthus retusus has never been reported. As a result of that, it is necessary toestablish the rapid propagation system of Chionanthus retusus Lindl.et Paxt. and toshorten the breeding cycle to furnish theoretical and practical evidence of theproduction of seeding.This test by Chionanthus retusu Lindl.et Paxt. embryo as explant, to analyzes theoptimal disinfection conditions to Chionanthus retusu Lindl.et Paxt. embryo, theoptimal ratio of mediumand the mixture ratio of hormone to get the complete plant.What’s more, this test uses aseptic seeding with top buds and the lateral bud stemsegments as material. With different proportion of hormones, we can get the completeChionanthus retusu regeneration and confirm the optimum conditions of regenerationsystem. Then, by using leaves of Chionanthus retusu Lindl.et Paxt. to induce callusand giving different concentrations of salt stress, we analize the physiologicalindicators in a certain period. The main results are of the following:1. The best conditions for disinfection of the embryo is:75%C2H5OH for30s;HgCl2for2min, the survival rate is94%; 2. The best rooting medium for embryo germination is:WPM+0.1mg·L1NAA+3.0mg·L1GA3+1.0mg·L16-BA;3. The bud sprouting of embryo medium is:WPM+1.0mg·L1GA3+1.5mg·L16-BA;4. The most suitable medium for the proliferation culturing of the Chionanthusretusu with top and the lateral bud stem segments is:WPM+1.0mg·L1GA3+1.0mg·L16-BA+0.5mg·L1TDZ;5. The most optional rooting medium of the Chionanthus retusu with top and thelateral bud segments is:WPM+0.1mg·L16-BA+0.5mg·L1NAA+0.5mg·L1IBA;6. The optional medium of the cotyledon to induce callus is:WPM+0.5mg·L1NAA+0.5mg·L12,4-D+0.1mg·L16-BA;7. The active content of soluble sugar, soluble protein, proline, MDA, SOD andPOD changes of Chionanthus retusus callus at different concentrations of NaCl stress(0、50、100、150、200mmol·L1) are as following: the content of soluble sugar andproline rises first and then dcreases in which the content of Chionanthus retusus calluswhich were treated by50mmol·L1NaCl were lower than the untreated group. Otherindicators, the content of the soluble protein rises first and then decreases. The contentof MDA increases rapidly, declines slowly in the midanaphase. The control groupdeclines continually. The activity of SOD is rising rapidly. The activity of PODdecreases significantly initially and then increases significantly later. While thecompared group has no significant changes with the treated group.
