Tree peony (Paeonia suffruticosa Andr.) is Paeoniaceae, Paeonia lactiflora, Sect.Moutan, which is fleshy root. Peony is a kind of excellent ornamental, medicinal andoily species. In this paper we used the bubil of ‘Luoyanghong’ where are in theInternational of Peony Garden as the research material, from tissue culture plants ofdifferent medium of morphological indexes and physiological indexes, cultureenvironmental conditions as three aspects were studied, in order to select the optimalculture medium and culture conditions. The results of the study were as follows:1. In five different culture mediums, which all can induce the bulbil of‘Luoyanghong’ to grow explants normally. MS medium’s and DKW medium’splantlets are growing well. The height of plantlets, the thick of stems, leaf area, leafnumber are better than the medium of1/2MS, WPM, LP. And the content of solubleprotein, soluble sugar, starch are lower than the medium of1/2MS, WPM, LP. Thegrowth vigor of plantlets is related to the nutrients condition of plantlets.2. In five different culture mediums, the chlorophyll content of thephysiological indicators in the plantlets is correlated negatively withanthocyanin contents. The content of anthocyanin of leaves is higher than otherfour kinds of mediums, while the content of chlorophyll is opposite. The leavesappear purple, they may are stressed by sugar.3. In five different culture mediums, the antioxidants (SOD, POD, PPO)activity of LP medium are higher than other four mediums, which showed thatculture plants of the LP medium are being forced to produce a series ofprotective response. MDA contents of LP medium are highest, PPO activity inLP medium rise so quickly that it causes increase of browning rate.4. The plantlets in five different culture mediums,‘Luoyanghong’ tissueculture plantlets’ browning rate and vitrification rate in changed temperature condition are lower than tissue culture plantlets in steady temperature condition,which showed that peony tissue culture plantlets are suited to the condition ofchanged temperature.In view of this, with the bulbil of ‘Luoyanghong’, we should choose MS orDKW, and we must provide changed temperature condition.
