Morinda citrifolia, also known as NONI, is a kind of famous tropical medicinal plants. Morinda citrifolia has become the hot spot of research and development and a new industry in Hainan with its good health care function. Construction of core collection of NONI can effectively improve its germplasm preservation and utilization efficiency, thus alleviating the increasing stress in germplasm preservation. But so far, no studies have been reported at home and abroad to construct core collection of Morinda citrifolia. In this study, biological traits, agronomic traits, SRAP molecular marker data were used to study the NONI core collection construction method. By comparing6kinds of system clustering method,4kinds of systematic sampling strategy,8kinds of sampling proportion within group,4ways of effective detection, combined with molecular markers and production practice, aimed to investigate the best clustering processing system, the optimal grouping principle, the optimal sampling proportion to tackle the mechanism of core germplasm construction of NONI. At the same time the result desire to construct the core germplasm and evaluate the quality, the genetic diversity and the germplasm selection rate of Morinda citrifolia core germplasm. The results will provide a theoretical basis for the conservation and utilization of germplasm of Morinda citrifolia, providing reference for the utilization of other tropical plants core germplasm construction in China, which has strong theoretical and practical significances. The main results are as follows:1. In the Euclidean distance, using the SAS software to analyze the traits data of Morinda citrifolia under6different cluster methods which including UPGMA, Ward’s method, Centroid linkage, Median linkage, Complete linkage and Single linkage WARD and compare the results. The results prove that the6systematic cluster analysis methods are all very good for clustering, with the UPGMA(AVERAGE)cluster method is the best, followed by the CENTROID linkage.2. Based on system clustering, comparing the four sampling strategies:proportional strategy(P), square root strategy(S), logarithmic strategy(L), genetic and diversity-dependent strategy(G),the result showed that genetic diversity proportion (G) had the best effects.3. Using the best systematic cluster method (AVERAGE) and the optimal sampling strategy (G), through the four efficacy data(MR:means of traits, SR:variance of traits, RR:Coincidence rate, CR:Variable rate) compared the8different groups of sampling ratio:5%,10%,15%,20%,25%,30%,35%and40%. It was found that the sampling ratio of 15%has the most significant effects. Initially constructed Noni core collection which containing18noni accessions.4. Further slected the initially noni core collection which contains18noni accessions constructed which through clustering in verage system, G sampling strategy, sampling ratio of15%in the group by SRAP data, which was constructed without apparent genetic redundancy.5. Combined with the reality of production practices need to artificially selected not elected to the good genes. HM121has strong cold resistance, cold resistance strains are cultivated noni dominant gene. The result of the amount of HX320is very high, is to cultivate high-yield strains of good genes. HN835drought resistance. These three lines artificially selected noni preliminary core collection, contains21noni accessions constitute the initial core collection of Morinda citrifolia.The most suitable method for constructing core collection of noni is using AVERAGE system cluster, G sampling strategy,15%of the sampling ratio within the group, based on the combination of molecular markers and production requirements constructed the core collection which contains21noni accessions. The reliability of core collection was identified.
