Endometritis is a postpartum disease of daily cow, this kind of disease is usually caused by pathogenic microorganism invasion, which injured the endometrial.Our experiment build a pathological mode of endometritis by cows, which intubating uterine horn by surgery operation, artificial noculating Staphylococcus aureus. On the basis of observing changes of the model, which include physiological index, blood index, uterine cervix secretion, histopathology and ultrastructure of uterine mucosa, We may probe into changes mechanisms of nuclear factor-κB (NF-κB), a kind of cell signaling pathways of inflammation in innate immunity, when cow endometritis infected in acute by Staphylococcus aureus.Six healthy adult cows were used to make molds, uterine horn intubated of aseptic condition and Staphylococcus aureus artificial noculated.3.5×109cfu Staphylococcus aureus was infused into cow uterus through the uterine horn intubation7days later after surgery operation. Observing and measuring body temperature (T), sphygmus (P) and respiratory rate (R), Collecting blood samples and uterine cervix secretion on0,3,6,12,18,24,36,48and72hours after inoculated. Counting statistics of white blood cell and neutrophil from blood samples. Differential counting cells from uterine cervix secretion samples.The experiment results indicate that:1. Body temperature (T), sphygmus (P) and respiratory rate (R) show a certain degree of rise after inoculated. There are a large number of secretion from vulva in3hours,6hours,12hours.18hours,24hours,36hours and48hours after inoculated. Secretions increase gradually, purulent and getting darker from3to24h, most purulent and sticky from6to18hour especially.Secretions become to thin and reduce gradually from24hours to48hours, while rarefied and clear72hours after inoculated. Microscopic examination shows that the number of PMN in Secretion has increased dramatically, PMN%has significantly higher levels (P<0.05).2. The sum number of WBC in peripheral blood also become significantly higher levels (P<0.05), as well as PMN (P>0.06).the number of WBC. PMN and PMN%reduce respectively in the end. Pathological observation results show that uterus mucosal epithelial cells falling off largely. PMN plenty infiltration, submucosa turning to thicker and uterine gland changing to anomalous. SEM indicate that the uterus mucosal epithelial cells fall off, fragmentary and scathing, Microvilli and cilium is damaged, fractured and disappear.3. Test results show that peripheral blood and secretions contain a higher β-defense element, C-reactive protein and cytokines after Staphylococcus aureus inoculated. The content of P-defense element, C-reactive protein, TNF-α. IL-8, IL-6and IL-1β became significantly higher levels (P<0.05), while fall back in the end. 4. Gene expression quantity of TLR2, TNF-a, IL-8, IL-6and IL-1β in WBC, which is collected from peripheral blood and secretions, has increased significantly higher levels (P<0.05), Gene expression quantity of TLR4appears higher, also.Thus it can be seen that NF-κB was induced by TLR2when infected in acute by Staphylococcus aureus. Endometrium was induced inflammatory response by target gene transcribed and inflammatory factor released.
