Leptocybe invasa adults from different seasons were chosen as the test object in this study. Exposure experiment and acclimation experiment were used to test L. invasa adults’resisitance ability to extreme temperature. Expression changes of insect protein were analyzed by two dimensional liquid chromatography and mass spectrometry. And some methods of bioinformatics were used to research the biological function of various distinct proteins.This study was to provide theoretical foundation for pest forecasting and control, and promote the development of L. invasa proteomics. The main results are shown as following.(1) The exposure experiment proved that L. invasa adults have a certain basal tolerance to extreme temperature, and compared with autumn, summer bodis have higher heat resistance but lower cold resisitance, autumn bodis were contrary.(2) Similar conclution was reached from the cold acclimation, besides, the result showed the induced tolerance of L. invasa adults to cold, and long-term exposure under non-lethal low temperature may cause chilling injure to insects.(3) Based on the result of exposure experiment,4℃2h and40℃2h were established as the induction codition for distinct proteins. Phenol extraction method was used to evtract the total insect proteins. Proteins expression of L. invasa adults under extreme temperature was analyzed by quantitative markers, combined with2D SCX/RP-HPLC and LTQ/Orbitrap-MS. There were339distinct proteins identified. Compared with normal temperature, insects under high temperature had67proteins were up-regulated, while32proteins were down-regulated; insects under low temperature had28proteins were u-regulated, while57proteins were down-regulated, there were53overlop between these two groups.(4) The biological function of90proteins were identified by database, there were heat shock proteins, and some other proteins about protein metabolism, energy metabolism and nuclease protection, such as Heat shock protein68, Heat shock70kDa protein congrate1,40S ribosomal protein S2, Elongation factor1-a1, Protein disulfide isomerase, Glyceraldehyde-3-phosphate dehydrogenase1, Cytochrome c-1, Histone H2A, DNA repair protein Rad23, etc..This study provides some experimental basis for learning the tolerance of L. invasa to extreme temperature and the biochemical mechanism, and some reference of theory and technology for carrying out the protemics of L. Invasa.
