Exploring Of Two Molecular Markers Associated With Pre-harvest Sprouting Tolerance In Spring Wheat In Inner Mongolia

Pre-harvest sprouting (PHS) seriously affect its economic value and end-use of quality, and PHS is a common problem in many wheat prosuction areas in China; therefore, breeding PHS resistence varieties become more and more important and it is the direct method solving PHS problem. Seed dormancy is a major factor that controls the PHS under wet weather condition. In this study, two STS markers (Vp1B3and Vp1A3) associated with PHS tolerance evoluated the genotypes of PHS tolerance in137red spring wheat in Inner Mongolia. The results showed that, three kinds of fragments (845bp,569bp and652bp) fragmens were amplified with STS markers Vp1B3in the137genotypes, and Vp1Bb and Vp1Bc are PHS resistant genotypes, wheareas Vp1Ba is PHS susceptable genotypes. the frequencies of the Vp1Bb,Vp1Bc and Vp1Ba in this set of germplasm were2.92%,81.75%and14.60%, respectively. In addition, a heterozygous Vp1Ba/VplBc is detected. Five kinds of fragments,725bp,774bp,733bp,765bp and762bp marked with the letters I, A, H, J and G,were detected with another STS marker Vp1A3in137genotypes. The frequencies of these fragments were33.58%,2.19%,42.33%,1.46%and20.44%, respectively, haplotype of G fragment is PHS tolerance genotype. Combining with the VplB3and VplA3markers, the efficiency of marker assisted selection for PHS resistant varieties was improved. The haplotype combination of Vp1Bc and VplAg was associated with more PHS Tolerance, a total of24genotypes were haplotypes combination of Vp1Bc and VplAg. Moreover, exploring the TaGAMyb polymorphic sites was detected in genotypes with different PHS tolerance, the results showed that polymorphic fragments were detected with the primer sets TaGAMyb-B.
