Historical wheat in China was not only rich genetic resources, but also are parental resources in wheat breeding. At present,the Pre-harvest-sprouting (PHS) in wheat is a serious problem in safe of wheat production and improving the quality of wheat, therefore, breeding PHS tolerance wheat has an urgent problem in wheat breeding. Marker-assisted selection in wheat breeding can shorten the time of breeding effectively. We selected Vp1A3and Vp1B3associated with PHS tolerance to evaluate PHS tolerance varieties in a set of historical wheat germplasm in Chinese. The results indicated that, Eight alleles of Vp1-A were detected, which were Vp1-Aα, Vp1-Ab, Vp1-Ag, Vp1-Aj, Vp1-Ak, Vp1-Ai, Vp1-Ah and Vp1-Al, with frequencies of28.04%,8.41%,2.80%,7.48%,3.74%,38.32%,8.41%and1.87%, respectively. In addition, the Anxuan2was the new type, named Vpl-Ar, compared with Vp1-Aα, which had a15bp (5CTT) deletion in position of2718bp-2732bp. Three alleles, which were Vp1-Bc, Vp1-Ba and Vp1-Bb, were detected by marker Vp1B3associated with PHS tolerance with proportions of44.86%,44.86%and9.35%, respectively. Combination with the result of Vp1A3, Vp1B3and germination index, five resistance genotypes were selected out, which were Anxuan2, Yannong15, Zhengzhou683, Zhengzhou742and Kequn. The haplotypes of Vp-1in these five cultivars were Vp1-Ar and Vp1-Bc; Vp1-Ah and Vp1-Bc; Vp1-Ab and Vp1-Bc; Vp1-Ag and Vp1-Bc; Vp1-Ah and Vp1-Bc.We also research the allelic variation in Ga20oxl in different PHS tolerance wheat. Compared with the sequence of Y14008, Ga20oxl-B of Zhongyou9507and Jingdong1had three SNPs, which all lead the change of amino acid; For GA20oxl-D, Jingdong1had three SNPs at the position of379bp,581bp and1101bp, respectively, and Wanxianbaimaizi had a SNP at positions of1161bp, these SNPs all lead the change of amino acid.
