| To find a good way to rapid and practical diagnostic method of bovine ketosis, a replacement samples of blood was tested by whoe milk and whey. The feasibility and reliability of whey instead of blood was examined. In this study,206cows was selected from a large-scale dairy farm in Guangxi Nanning, modified salicylaldehyde colorimetry was used to detect the ketone body content of blood plasma, whey146cows, to detect the ketone body content of blood plasma and whole milk in60cows. Plasma lvels of ketone body being more than5mg/dL was diagnosed as cows with subclinical ketosis, while more than10mg/dL as clinical ketosis. Results:in146cows,10cows were diagnosed with clinical ketosis,13cows were diagnosed with subclinical ketosis,11cows were diagnosed with suspected ketosis,112cows were diagnosed with health, ketosis incidence was15.75%. according to milk ketone,28cows were diagnosed with clinical ketosis,58cows were diagnosed with subclinical ketosis,33cows were diagnosed with suspected ketosis,only17cows were diagnosed with health, ketone disease incidence was58.9%2. In60cows’study, There was no cows diagnosed with clinical ketosis and suspected ketosis, but3cows were diagnosed with subclinical ketosis,57cows were healthy, so the disease incidence is0.by milk ketone:2cows were diagnosed with clinical ketosis,19cows were diagnosed with subclinical ketosis,22cows were diagnosed with suspected ketosis of cattle head, only17healthy cattle head, ketone disease incidence was35%.3. The correlation of whey ketone content and serum ketone content is0.61, higher than the correlation (0.26) of milk ketone content and serum ketone content. ROC curve:whey ketone threshold is10.96mg/dl while serum ketone threshold is5mg/dl,the area under the peak is0.779; whey ketone threshold is24.17mg/dl,while serum ketone threshold is5mg/dl, the area under the peak is0.892.5. With the cut point of serum ketone10mg/dL, serum ketone’s area under the ROC curve is0.995in95%confidence, whey ketone’s area under the ROC curve is0.797. Whey ketone and serum ketone’s r value is0.613, and cubic equation R2value maximum (R2=0.495) during SPSS’s best relationship function screening. Conclusion, the average plasma contents of ketone body in146cow was6.28±9.57mg/dL, while that in whey is14.2±8.25mg/dL; the average milk ketone content in60cows was9.29±4.79mg/dL, the average contents of plasma ketone was2.44±2.23mg/dL. It was confirmed that It could be used milk serium to determined ketone body in diagnosis of bovine ketosis. The equation was found between whey and plasma ketone bodies (serum=y, whey=x):y=0.01x~3-0.024x~2+0.604x-0.304. |
