Objective To study the effects of the complex type oligosaccharides on the growth,reproductive ability related reproductive hormones, sperm motility in the most suitable mating,testicular antioxidant content, organization structure and the expression of DDX4protein inmale mice.Method Thirty-six21d female mice and thirty-six21d male mice were randomlydivided into three groups: control group (NC), chitosan oligosaccharide group (COS),complex type oligosaccharide group (CTO).There were24mice in each group,12femalemice and12male mice were selected for experiment. Seven weeks of the mice wererandomly mated for five days after the experiment commenced, when the weaning threeweeks of the mice and sexual maturity six weeks of the mice, we collected issue, blood andother relevant samples. The morphological structure of testicular tissue, using HE stainingmethod, reproductive hormones and testis antioxidative capacity index were determined bythe ELISA kit; determination of epididymal sperm viability is determined by blood cell countmethod, Expression of Western protein was detected by DDX4blotting method.Results (1) Compared with the NC group and the COS group, the average body of eachweek were significantly increased in the CTO group after the experiment commenced(P<0.05). The change of weight among all trial groups of F/G showed the same trend.(2)Three weeks of the mice, compared with the NC group and the COS group, liver index,spleen index were significantly increased in the CTO group (P<0.05); Six weeks ofmice,compared with the NC group, testis index were significantly increased in the CTO group(P<0.05); Twelve weeks of the mice, compared with the NC group and the COS group, liverindex, spleen index, epididymis index were significantly increased in the CTO group (P<0.05),testis index of CTO group and NC group, COS group compared to the highest, the significantdifferences between groups.(3) The content of T (testosterone) in the blood serm was thehighest in the CTO group, and was lower in the COS group, and was the lowest in the NCgroup, there were significant differences between each group (P<0.05),3,6,12weeks of ageshow the same trend.(4) Six and twelve weeks of the mice, compared with COS group andNC group, the number of sperm respectively increased by7.17%,8.19%and18.49%,12.57%in CTO group (P<0.05), compared with COS group and NC group, the number ofspermatogenic cells were increased by7.87%,8.87%and43.79%,13.54%in CTO group (P<0.05), compared with COS group and NC group, the number of leydig cells wereincreased by5.37%,8.72%and6.75%,10.94%in CTO group (P<0.05).(5) SOD (superoxidedismutase), GSH-Px (gutathione peroxidase) content was the highest and MDA(malondialdehyde) content was the lowest in the testis, there were significant differencesbetween each group (P<0.05), each weeks of age show the same trend.(6) Compared with NCgroup, the percentage of the epididymis moves rapidly forward sperm (a) were significantlyincreased in COS group and CTO group, a significant difference between groups (P<0.05);compared with NC group, the epididymal sperm motility were increased with significantdifference in COS group and CTO group.(7) Compared with NC group, the expression oftesticular DDX4protein were increased significantly in CTO group (P<0.05); at the age of12weeks and3weeks of age showed the same trend.Conclusion complex type oligosaccharide can promote the growth of male mice andimprove the reproductive ability of male mice. |
