Acyrthosiphon solani is the piercing sucking pest of soybean, which the resistance and damage are increased year by year according to the results of field survey in recent years. In order to understand the odorant binding protein mechanisms of Acyrthosiphon solani and explore the new target for prevention, we cloned an odorant binding protein gene named AsolO3P7by real-time PCR (RT-PCR) and RACE (AsolOBP7stands for odorant binding protein7gene from Acyrthosiphon solani in the following)(Accession Number:KF813022). The relative expression of AsolOBP7at mRNA level in every developmental stage and different tissue were analyzed by RT-PCR. One-way anova was analyzed by DPS. Antenna and cornicle of Acyrthosiphon solani were observed by scanning electron microscopy. The results showed that AsolOBP7cDNA is735bp in length, encoding155amino acid, the predictd molecular weight (MW) was17.52kD, and the isoelectric point (pI) was8.78. The predicted N-terminal hydrophobic region containing31amino residues displayed the characteristic features of a signal peptide. From the results of RT-PCR, it can be seen clearly that AsolOBP7expressed in different tissues of Acyrthosiphon solani, specially in antenna tissue, the expression of AsolOBP7got a peak, and showed significantly higher than other tissues (P<0.01). There were lower expressions in head (excluding antennae), legs, thorax, abdomen and wings, and it was found no significant difference between head and legs, and the same result during thorax, abdomen and wings (P<0.01). In addition, AsolOBP7was expressed at different developmental phases of Acyrthosiphon solani, it was detected lower expressions at lth、2th、3th instar and winged aphids period, and not significant difference (P<0.01). But it increased at4th instar, there is the highest expression during wingless aphids period, which has extremely significant difference with other period (P<0.01).In addition, there are primary rhinariun、secondary rhinarium、sensilla chaetica、sensilla campaniformia and non-peripheral scolopidia on antenna of Acyrthosiphon solani. The cornicles are cylindric and get longer with age. The results suggest that AsolOBP7is mainly in the antenna tissue and the most active in wingless adult aphid period.AsolOBP7is closely related to the olfactory behavior of antenna and may play a key role in the olfactory activity of wingless adult aphid period, which laid the foundation of looking for new control strategies.
