Atrijuglans hetaohei Yang(Lepidoptera: Oecophoridae),is one of the most destructive pests of walnut fruit.This study aims to understand the antennal morphology and ultrastructure of the moth A.hetaohei,and in order to investigate the biological significance of the behavior that when crawling or resting,adultA.hetaoheihas the habit of raising and swinging its hindleg.The ultrastructures of antenna and leg in both male and female adultwere studied using scanning electron microscopy.At the same time,to understand the mechanism of antennal olfaction in A.hetaohei,the RNA of the antennae of A.Hetaohei was extracted,transcriptome sequencing was performed and the olfactory related genes were selected and identified.The main results are as follows:1.The results showed that the antenna was filamentous comprising scape,pedicel and flagella.The flagellum consists of 45 and 51 segment in male and female,respectively.Nine types of sensilla on the surface of antennae were observed: sensilla trichodea,sensilla chaetica,sensilla auricillica,sensilla coeloconica,sensilla styloconica,sensilla squamiformia,B?hm bristles,sensilla basiconica and campullacea sensilla.Sensilla trichodea,sensilla auricillica and sensilla coeloconica had two subtypes,i.e.,sensilla trichodea I and sensilla trichodea II,sensilla auricillica I and sensilla auricillica II,sensilla coeloconica I and sensilla coeloconica II.The distribution and quantity of each sensillum,were different in each segment of antennae.2.The scape and pedicel of antennae were covered with scales,whereas the dorsal and lateral sides of each flagellar segment,except for the last segment,were covered with scales.There were holes,ridges,ribs,microribs structures on the scales.Holes can be divided into seven types: single hole,double,three,four,five,six and seven holes.There are certain rules and differences in the arrangement and abundance of holes in the scales of antennae.3.The results showed that fore leg,middle leg and hind leg of the male and female were all walking leg.There was no significant difference in sensillum,claw and setal pulvilli of hind leg,fore leg and middle leg.But,hind leg was much stouter than the fore leg and middle leg,and hind tibia was covered with more lanceolate scales.There were four main types of sensilla in both: sensilla squamiformia,sensilla chaetica,sensilla trichodea and B?hm bristles,leg scales could be roughly divided into five types,including narrow leaf scale,broad leaf scale,round leaf scale,lanceolate scale and conical scale.4.We identified 83 putative olfactory proteins: 29 OBPs,26 ORs,9 CSPs,19 IRs.This study described the ultrastructures and characteristics of the sensilla and the holes in the scales of the antennae of A.hetaohei,several olfactory genes were screened out,it provides a basis for further understanding of its chemical communication system and the development of new green prevention and control technologies;also clarified the morphological types of fore leg,middle leg and hind leg,and described the ultrastructure of sensillum,scales,claw and setal pulvilli on the leg,all of which provided a morphological basis for the further study of the biological function of the raising and swinging hindleg habit of A.hetaohei.
