Tomato is one of the most important vegetable crops,it is the main cultivated vegetable crops in our country. In China, the planting area of tomato has expanded each year, however, with the continuous improvement of the production, the tomato diseases is becoming more and more serious, among them, tomato yellow leaf curl virus disease has been one of the important diseases. At present, the disease is increasing year by year in China, from south to North tends to spread rapidly, causing tomato yield reduction even no production in some areas. For the control of this disease, there is no particularly effective method or agent, therefore, screening of resistant materials, disease resistance breeding has become the priority among priorities of the current work.Through the way of molecular marker in this experiment,researching the resistant materials at the level of genes of tomato yellow leaf curl virus disease, building the resistance, susceptible population, establishing the AFLP marker system and the SSR marker system, using each generation material screening and resistance to tomato yellow leaf curl virus Ty-2gene to linkage marker, lay the foundation for resistance to tomato yellow leaf curl virus disease breeding.The main research results of this study are as follows:(1) The experiment use ’CLN2498D’ which is resistable to tomato yellow leaf curl virus as male parent,’ZaoFen2’which is susceptible materials as female parent, obtain hybrid Fi generation, F1generation self to obtain F2generation, bemisia tabaci inoculation virus, building the resistance, susceptible population, and doing resistance identification to each generation of tomato. The results of identification show that:all the Fi generation showed resistance; actually segregation ratio of F2groups was3.09:1, consistent with the segregation ratio of3:1, by χ2test, consistent with single gene dominant inheritance(2) Male, female, F1generation and F2generation as the research object, using AFLP and SSR two kinds of markers, markers linked with Ty-2gene. Through the screening of256pairs of AFLP primers and30pairs of SSR primers, we get5AFLP markers and2SSR markers linked to Ty-2gene, the one marked with E02M11is5.8cM away from target gene, and another3markers are within10cM range.(3) Mark E02M11is used for germplasm screening, of which12tomato materials contain the marker, providing the basis for resistance to yellow leaf curl virus breeding.
