Wheat stripe rust is one of the important airborne fungi diseases, which distributed widely in the wheat growing areas of the world, an particularly in China occurs in severe damage, and seriously affected the yield and quality of wheat and threat to production safety of China’s wheat. The use of resistant varieties to prevent wheat stripe rust is the most cost-effective measures. Because of wheat varieties resistant to stripe rust low genetic diversity, and stripe rust are highly toxic physiological variability and the new generation lead pathogenic type species emerging, and resistant varieties of existing content easily be overcomed by new races and " lost " the original disease resistance in the short time.. Therefore, continue to explore and study new resistance genes, and use them in breeding for disease resistance, foster multi-gene varieties with durable resistance polymerization characteristics is particular important. Each wheat-growing areas of ecological differences, natural selection has preserved a rich peasant varieties resistant genes and allelic variation, which is a very valuable genetic resource library, and screen it to identify new genes for resistance to stripe rust to rich our country wheat stripe rust resistance gene resource pool has important theoretical and practical significance.According to the feature that adult resistance to stripe rust of landraces small Hongmang, using the conventional hybridization analysis methods, we can get Fl, F2and BC1generations through small Hongmang and susceptible variety Taichung29hybridization, selfing and backcross. In the field of adult vaccination stripe rust races CYR32, disease resistance were identified and statistical analysis of their parents and offspring. The results showed that small Hongmang for CYR32adult resistance to stripe rust is controlled by a recessive gene.Landraces old white wheat and white lines Mans all have the feature that growth period resistance to stripe rust. We have got F1, F2and BC1generations through building hybrid genetic population of old white wheat and susceptible variety Taichung29. In greenhouse seedling inoculation stripe rust CYR33, and CYR32rust in its field in reciprocal cross inoculation groups of conduct resistance identification and statistical analysis of their parents and offspring. Results show that the growth period resistance of old white wheat to CYR32and CYR33is controlled by one dominant gene and belong to nuclear genetic.Building resistance pool and susceptible pool by F2genetic population that from genetic analysis. Using SSR technology randomly selected560pairs of primers on21wheat chromosomes, then genomic DNA polymorphism analysis to the parents white lines Mans and Taichung29, resistance pool and susceptible pool. The result show that, the primer wmc658has polymorphism between parents and resistance susceptible pool. Using primer wmc658and the genomic DNA of230F2population plants for PCR amplification and PAGE analysis, we got wmc658linked with target genes of genetic distance of6.9cM, the resistance to stripe rust disease gene is located on chromosome2AL.
