This study collected three dairy farm milk samples and the calf blood samples. Used the colostrummeter, Brix refractometer and serum protein refractometer to evaluate the quality of colostrum and the content of serum protein on site, and used laboratory methods HPLC to determine colostrum samples, serum samples1gG. And to establish relations between Brix refractometer and colostrometer. The failure rate data of Taian calf passive immunity was set up by collecting calf blood samples in different feed systems. Setting relations between different feed time、different feed amount and calf passive immunity. The study included three parts:1.The study of calf passive immunityThough3dariy farms research,the passive immunity failure of them were57.14%(farm1),37.14(farm2),14.81(farm3),respectively.The result showed that different feed systems of Newborn calves played a very important role in obtaining passive immunity for calves. The key to improve calf passive immunity is to set up and perform good colostrum procedures(colostrum quality, feed time and feed amount).2.The study of colostrum quality assessment technology.Collecting the colostrum sample squeezed first time after the cow produced calves in different dairy farms. Using Brix refractometer to measure the Bix value, and to determine the milk composition (Fat,Pro,Lact、TS) of colostrum. Establish the relationship between the Bix and IgG,milk composition and IgG. Compared with HLCP laboratory method, using Bix refractometer and colostrometer to evaluate the accuracy of colostrum quality on site. The test results show that the IgG and sugar value, milk protein, total solids have a good relationship3.The study of feed time, feed amount for calf passive immunity.By feeding experiments showed that feeding the same quality colosturm on the same time when the first fed volume is4L,the serum total protein values is6.28g/dL,which thepassive immunization level is the highest;however,when the first fed volume is4L,the IgG concentration is19.12mg/mL,which the passive immunization level is the highest.the differences between the groups was not significant(P=0.95);at certain feeding volume of feeding colostrum,determined by the protein was measured using a refractometer,when the first fed time is4h,calf passive immunity is higher,and less passive immunization failure of the indicidual;however when determine IgG concentration by HPLC,the passive immunization of calves level is17.982mg/mL when first fed time is2h,the difference between the groups was not significant(P=0.27).
