This study focused on Nocardiopsis, one genus of rare actinomycetes, isolated from south edge of the Takelimakan desert, Xinjiang. By the HP-20macroporous resin absorption and elution by different ration of acetone with water, a fingerprint comparision model was established by HPLC between the blank media and the elution of secondary metabolites. In addition, the secondary metabolites from different strains were compared. On the basis of HPLC profiles, the following three research works were carried out:Firstly,6Nocardiopsis strains from south edge of the Takelimakan desert, Xinjiang, with activity against Staphylococcus aureus, were cultured in the same condition and the same media. After the same treatment, a fingerprint was set up on the purpose of comparisons between the blank culture medium and secondary metabolites; this work was established for directly finding new antibiotics.Secondly, based on the fingerprint model, the strain R2A14A-1, which may produce amicoumacins, was found. After fermentation, isolation and purification, FFF-13, a di-hydrogen isoconmarin compound, with inhibitory activity against Staphylococcus aureus was obtained from the strain R2A14A-1. After spectral analysis and the literature comparison, the compound FFF-13was determinded as amicoumacin B. The chemical structure was show in figure1. As best of our knowledge, this is the first report Amicoumicn B was produced by Nocardiopsis sp. Fig.1Structure of amicoumacin BThirdly, the strain R2A14A-1was classified as Nocardiopsis aegyptia by morphology and cultural characteristics, the optimum growth condition and NaCl tolerance, physiological and biochemical characteristics,16S rRNA sequence analysis and (G+C) mol%. The results showed that it shared99.14%with Nocardiopsis aegyptia DSM4442T (AJ539401) of16S rRNA gene sequences, Finally, as a result of the study, the strain R2A14A-1was belonged to the genus Nocardiopsis.
