In order to investigate the distribution of GTHR and expression of GTHR mRNA during estrous cycle in the reproductive tissues in bitches, the experiment made the ovary, uterus and oviduct of bitches of follicular stage and CL phase stage as the research object. According to the study, using the method of immunohistochemical SABC and in situ hybridization, to localise the FSHR and LHR and detect the semi quantitative expression of GTHR mRNA during estrous cycle in the reproductive tissues in bitches. What the results shows as follows:1、Both of the follicular stage and CL phase stage, the distribution of positive cells of FSHR and LHR was observed in ovary and different follicle stage. The most FSHR positive cell amount is the mature follicle(178.6±3.026) which is in the follicular stage, and significantly higher than other positive cell amount; The most LHR positive cell amount is the antral follicle (104.0±0.9129), which is in the CL phase stage, and significantly higher than other positive cell amount. Both of the follicular stage and CL phase stage, the distribution of positive cells of LHR was observed in oviduct, the distribution of positive cells of FSHR was only observed in the follicular stage, in the CL phase stage had no reaction.2、The hybridization signals of FSHR mRNA and LHR mRNA were observed in the reproductive tissues both of the follicular stage and the CL phase stage, with using the method of in situ hybridization; The result of semiquantitative expression shows that, in the follicular stage, the relative amount of FSHR mRNA in the ovary is the most (120.408±1.768), and significantly higher than oviduct; The relative amount of LHR mRNA in the uterus is the most (117.032±0.151), and significantly higher than ovary and oviduct; In the CL phase stage, the relative amount of FSHR mRNA of ovary is the most(102.006±0.352), and significantly higher than uterus and oviduct; The relative amount of LHR mRNA of ovary is the most (109.298±1.022), and significantly higher than uterus and oviduct.
