| Vitis Amurensis is an important wild resource in China. There are a variety of metabolites in V. amurensis, such as f lavonoids, tannins, phenylpropanoid and so on. In this paper, we studied the transcriptional regulation factor in the process of Anthocyanin Biosynthesis.SDS method, Trizol method and modified CTAB method were used to extract total RNA in the skin and leaves of V. amurensis in this experiment. The results show that the total RNA stripe is completed in the skin and leaves of V. amurensis by modified CTAB method. The fluorescent bringhtness of28s rRNA and18s rRNA is close to2:1. OD260/OD280values is between1.8~2.0. The purity and the extraction amount of the extracted total RNA by modified CTAB method are the highest.The full-length cDNA sequences of transcriptional regulation factor Vamybs genes in mature skin of Vitis amurensis were cloned by RT-PCR and SMARTTM ACE. The analysis of informatics biology show it encodes171amino acids. The molecular mass of cDNA sequences is19.47kDa. The isoelectric point is6.15. Instability index is30.56. which indicated that this gene protein is stable protein. Its similarity with Vitis vinifera and Hybrid grapes are all99%by multiple sequence alignment. The results of molecular evolution tree showed that the genetic relationships of Vamyb2with V. labrusca and Hybrid grapes are closer, but its genetic relationships with Antirrhinum and Arabidopsis are longer. The gene don’t have a signal peptide, but with hydrophilic and hydrophobic; The major structure components of protein in secondary structure mainly were α-spiral and irregular curl. As the same time, the gene protein do not have transmembrane domain, the function mainly is in outside of membrane. |
