Vitis amurensis grape berries can be used to make wine, is the main raw material for the wine industry, the key factors affecting the quality of the wine grape peel color, and the color of grape skins in turn depends on the composition and content of anthocyanins, in anthocyanin biosynthesis, in the process of anthocyanin biosynthesis, F3H gene is the key gene of flavanone and anthocyanin metabolic pathways.The present study is of Vitis amurensis ’Shuangfeng’ cultivars were successfully cloned cDNA sequence F3H gene and it’s with bioinformatics tools analysis, using Fluorescent Quantitative PCR Technology analysis of seven different periods in Vitis amurensis fruit maturation in the process of F3H gene expression level,and cloned the gene promoter, and predict its cis-acting elements.Experimental results show:1. VamF3H gene a total length 1398 bp, ORF is 1092bp, capable of encoding 363 amino acids. A molecular weight of 40.81KDa, the isoelectric point (PI) is 5.37. It does not have a signal peptide and the transmembrane domain, the hydrophilic protein belonging instability. The amino acid sequence of Vitis vinifera (FQ389950), muscadine (KF040970) and the American vine (JX087441) like F3H genes has higher homology.2. Real-time quantitative results:F3H gene in the Vitis amurensis skins maturation process is different the expression level of in each period, F3H not expressed in turn color the early stage, late F3H expression with the maturity of fruits increased gradually increased, ripe stage and decreased significantly.3. F3H gene start subsequence with typical CAAT and TATA box, in addition there are a number of light response response element, hormone response element, stress response elements such as that of the gene transcription and expression may be influenced by light, drought, hormones and other external environment, push measure it is stress related promoter gene promoter.
