Ligustrum lucidum Fruit (LLF) is a traditional Chinese medicine of nourishing liver and kidney, Selenium is an essential trace element for human beings. modern pharmacology studies showed that there are many similar biological efficacy between them, such as hepatoprotective effect, immune regulation, anti-oxidation and anti-ag ing effects. Therefore, we did experiments for Ligustrum lucidum Fruit to study biotransform selenium and quality evaluation of se-enriched Ligustrum lucidum Fruit(Se-LLF), then providing a thesis to develop a new kind of crude drug or se-enriched raw material. All these efforts are for the further exploitation of LLF.The research content includes:Testl Effects of selenium on the main nutritional and active components of LLFIn the full-bloom stage of Ligustrum lucidum,we respectively sprayed sodium selenite solitions with the concentration of0,100,400,500mg/L to the leaves of LLF at50ml/M2,the blooming spray was conducted for2times.Finally,We got four different se-enriched Ligustrum lucidum Fruit products.Then the contents of selenium, the nourishment compositions and main biological active ingredients of four kinds of LLF were determined. The nourishment compositions of LLF include moisture, ash, crude fiber, crude fat, crude protein and nitrogen free extracts, main bioactive components are Polysaccharide, flavonoids and oleanolic acid. The result showed that:the contents of selenium, crude protein and polysaccharide in Se-LLF increased significantly than LLF;the contents of oleanolic acid and crude fat declined slightly; the contents of total flavonoids had not changed. Components contents of Se-LLF at the concentration of400mg/L are:Se:1.88μg/g; crude protein:8.34%; polysaccharide:4.89%; flavone:0.52%; oleanolic acid:29.19mg/g. It suggests that the effect of improvement on crude protein and polysaccharide is the best, thus400mg/L Se-LLF can be used as the standard of Se fertilization.Test2A tentative exploration in the enriched area of selenium in Se-LLFThe results of Test one indicated that contents of protein, amylase and oleanolic acid in Se-LLF were varied, compared with LLF. We speculate that the reason of contents change is Selenium combined with the above components where probably Selenium enriched area is. To study the distribution regularity of selenium in Se-LLF, four different concentration of ligustrum lucidum fruit components, as Selenium enriched area, including ligustrum lucidum fruit powder, protein, amylase and oleanolic acid, were detected respectively. The results showed that Selenium content in ligustrum lucidum fruit protein was the most, accounted for over30%of the total amount of Selenium, and the level was as high as8.17μg/g. Selenium content in amylase was about1.15μg/g, accounted for3%-6%of the total amount of Selenium. And it was extremely lower than in protein. Oleanolic acid contained Selenium0.17μg/g, was the least compared with the others. It indicated that the major Selenium enriched areas in Se-LLF were protein and amylase, in which protein dominant.Test3Establishment and analysis of Se-LLF fingerprint technologyIn this experiment, the thin layer chromatography, infrared spectroscopy and ultraviolet-visible spectroscopy were respectively used to identify the distinction between LLF and Se-LLF.Thin layer chromatography mainly inspected alcohol extract ingredients and analyzed pharmacopoeia quality index of oleanolic acid in the LLF and Se-LLF. The infrared spectroscopy was established to investigate the diatingction of protein, polysaccharide and oleanolic acid between LLF and Se-LLF. The structures of total flavonoids were analyzed by ultraviolet-visible spectroscopy between the LLF and Se-LLF. The result shows that:Firstly, through thin layer chromatographys analysis, there was no change in chemical components and contents.Secondly, infrared spectroscopy results of the LLF and Se-LLF have subtle differences, the lack of peak and peak displacement are found which means that Selenium affected the structure composes of protein, polysaccharide and oleanolic acid and probably spectrum effect of selenium enriched area. Finally, the ultraviolet spectrum shows that the structure of the flavonoids in LLF and Se-LLF are the same proves that selenium has no effect on the flavonoids in LLF.
