Bacterial fruit blotch of melons and water melons caused by Acidovorax citrulli. is a world wide disease that causes serious damage on the melons.water melons and cucurbits plants,it has been reported that this disease can cause serious damages.now copper is still an dominating bactericide for bacterial fruit blotch of melons.due to the microorganism is well-adapted and mutable, the resistance of bacterium have been enhanced with the copper being widely used,which bring about Stronger pathogenicity.Through studying the heavy metal detoxification mechanism of pathogenic bacteria to know pathogenic mechanism will contribute to the effective prevention of Bacterial Fruit Blotch.Taking wild type strain Aac5that was isolated from watermelon for example and using the method of homologous recombination, we constructed the mutant strain Aac5△CueO which was verified by PCR and southern blot.The sulfate sensitivity,formation of EPS,Pathogenicity,HR on the non-host plant,the ability of growth, biofilm and motility of the mutant Aac5△CueO and Wild type Aac5was compared.The results are as follow:(1)The mutant strain Aac5△CueO is sensitive than Wild type Aac5,we can draw a conclusion that CueO genes is associated with resistance of copper.(2)The Pathogenicity of mutant weakened on melon and the formation of EPS decrease.HR on the non-host plant, ability of growth, biofilm and motility are no significant difference between mutant and wild type.
