Preliminary Screening On Balancing Blood Sugar Of Different Components In Citrus Changshan-Huyou Pulp

The Citrus changshan-huyou is a special kind of varieties of citrus in China, and it contains a lot of carbohydrates. The present study showed that Citrus changshan-huyou pulp was rich in active ingredients, which could balance blood sugar, but the ingredients roles were not clear. In this paper, the Citrus changshan-huyou pulp was separated to five components (light petroleum fraction, methylene dichloride fraction, ethyl acetate fraction, n-butylalcohol fraction and water fraction) with different polarities, and the main material in five components were tested. Then the fractions which could increase the glucose uptake in3T3-L1adipocytes were selected. In order to provide the reference for diabetes animal experiments, the five components were took to the acute toxicity studies on the normal mice. Finally through the diabetic mice model, five components which could balance blood sugar were selected. The results of the study are as follows:The Citrus changshan-huyou pulp was separated to five fractions, and the water fraction accounted for88.96±2.34%. The Citrus changshan-huyou were rich in hesperidin and naringin which accounted for1.59±0.25mg/g and3.74±0.38mg/g respectively. Flavonoids were mainly distributed in the ethyl acetate fraction, and a small amount of flavonoids distributed in the methylene dichloride fraction and n-butylalcohol fraction. Limonin compounds mainly refered to limonin and nomilin, which mainly existed in methylene dichloride fraction, ethyl acetate fraction and light petroleum fraction. Carotenoids of Citrus changshan-huyou pulp most existed in light petroleum fraction, and the rest existed in methylene dichloride fraction. Finally the carbohydrate of the Citrus changshan-huyou pulp mainly existed in the water fraction, and a few carbohydrate existed in n-butylalcohol fraction.This part was through the3T3-L1adipocytes model to screen components which could balance blood sugar. With the ethanol extract of the Citrus changshan-huyou pulp as experiment material, the experiment proved that the ethanol extract could significantly increase the glucose uptake in3T3-L1adipocytes. But when the concentration reached40mg/L, the effect of ethanol extracts reduced with the increasing of the ethanol extract concentration. The Citrus changshan-huyou pulp was separated to five fractions with different polarities, and the light petroleum fraction, methylene dichloride fraction, ethyl acetate fraction and water fraction significantly increased the glucose uptake in3T3-L1adipocytes. When the concentration was0~50mg/L, light petroleum fraction and water fraction significantly increased the glucose uptake in3T3-L1adipocytes with the increasing of concentration. When the concentration was50mg/L, the effects of light petroleum fraction and water fraction was the highest. And the effect of water fraction was higher than that of the light petroleum fraction. When the concentration was50~100mg/L, the effects of light petroleum fraction and water fraction were falling with the increasing of concentration. When the concentration of methylene dichloride fraction and ethyl acetate fraction was0~100mg/L, methylene dichloride fraction and ethyl acetate fraction significantly increased the glucose uptake in3T3-L1adipocytes with the increasing of concentration. When the concentration was30mg/L, the effect of n-butylalcohol fraction was remarkable. The results of the interaction between various fractions accorded that the effects of each fraction with other fractions were lower than the effects of the sum of them.The acute toxicity studies of five fractions showed that five fractions do not have vious toxic effects and death of mice. The weight increased normally when the dosage was2.4g/kg. The general behavior, movement function of the mice compared with normal control group was no significant difference. The breathing, coat, color, eyes, ears, micturition were all right. These showed that under the metering, five fractions of the Citrus changshan-huyou pulp had no effect on the physiological of the normal mices. So the five fractions had no effects of normal growth on the normal mices when the concentration was below2.4g/kg. The next experiment on the diabetes mice could be carried out.This part was through the diabetic mice model to screen components which could balance blood sugar. During the delivery, the weight gradually gained in mice. Compared with model control group, the water fraction of high dose could balance blood sugar in the diabetic miceIn summary, the Citrus changshan-huyou contained a lot of components (flavonoids, limonin, Carotenoids). The five fractions could significantly increase the glucose uptake in3T3-L1adipocytes. Compared with model control group, the water fraction of high dose could balance blood sugar in the diabetic mice. This study could provide references for screen some compositions which could balance blood in the Citrus changshan-huyou pulp.
