UPBEAT1(UPB1) is a transcription factor, which regulates the balance between cellular proliferation and differentiation from the Arabidopsis root. It can directly regulates the expression of a set of peroxidases that modulate the balance of reactive oxygen species (ROS) between the zones of cell proliferation and the zone of cell elongation. Studies have shown that disruption of UPB1activity destroys this ROS balance, leading to a delay in the onset of differentiation. This pathway functions independently of auxin and cytokinin plant hormonal signaling.In our research, we find UPB1-Likel and UPB1-Like2genes which are the homologous genes of Arabidopsis UPB1, by bioinformatics method, and we have cloned this two genes. We get a417bp CDS of UPB1-Likel and a408bp CDS of UPB1-Like2. The semi-quantitative PCR showed higher UPB1-Like gene expression in poplar stem and root volume, and it suggests that the genes may have important functions in poplar stem and root volume. Transient expression showed that poplar UPB1-Like gene location on the cell membrane and nucleus.To delve into its biological function, we use the Gateway cloning system successfully constructed of poplar UPBl-Like overexpressed genes (35S:UPB1-Like-GFP) and inhibit the expression vector (RNAi-Likel2-pGWB2) and inhibition of expression in Arabidopsis and poplar to get20mutant strains. The results showed that the morphology compared with wild-type, over-expression of the gene UPB1-Like poplar stems short and thin, short section spacing, whereas inhibition of poplar stems gene expression UPB1-Like long, thick, long section spacing. In summary, UPB1-Like genes affect the growth and development of poplar stems and roots. Especially for the accumulation of its growth regulation plays an important role. Therefore, the study for high grade to cultivate fast-growing forest tree varieties of production practice significance.
