Factory Plate Method For Cultivating Cordyceps Militaris

Cordyceps militaris was a famous medicinal and edible fungi. The suiTable spawn cultivated in local place and the suiTable mother culture medium, cultivate medium and conditions of cultivation in box played an important role in the cultivating of Cordyceps militaris. In this experiment, choose the spawn suite cultivate in Shanxi, optimization on Mother Culture Medium (carbon source, nitrogen source and pH), cultivation in fermentation tank (temperature, pH, liquid volume and cultivating time), cultivating medium (main material, accessories and nutrient solution adding) and cultivations conditions (inoculum, temperature,light time and air relative humidity). The result laid a foundation for factory cultivation. The results were as follows:1. Through a filter in the four spawns,we find the best spawn for factory cultivating in Shanxi: Cordyceps militaris RZ-1, provided by Mushroom science and technology in Shanxi agricultural university and Shandong Rui ZhiSheng service centers and Biological Technology Co.Ltd.2. The results shown that the best mother culture medium was potato200g, glucose(or sucrose)20g, agar6g, MgSO41g, KH2PO42g, beef extract20g, pH5.5～6.6, water1000ml.3. Mycelial biomass of Cordyceps militaris grown in100L fermentation tank were maximl through single factors and orthogonal test under the following conditions:temperature was22℃, initial pH was6.0, liquid volume was80L/Tank, culture time was3d.4. The results shown that the best medium formula, Wheat300g, Pupa powder15%, Liquid ratio1:1.7, Nutrient solution:water510g, glucose3.6g, KH2PO40.36g, MgSO40.2g. The best cultivating conditions: inocum20%, temperature22℃, light time10h, air relative humidity.5. Using factory cultivation, the utilization of the culture room was improved500%, The yield improved20%, cultivation times improved from2～3times to10times per year.
