| Pax7gene is an important transcription factor, which can regulation proliferationand differentiation of skeletal muscle satellite cells. The genetic mutation studies ofPax7gene mainly focused on humans and mouse, however, the studies of Pax7genewere few in the livestock and poultry, and no study of the Pax7gene about the geneticvariation in the chicken was reported.We used DNA pool sequencing method to scan the SNP sites of Pax7gene andPCR-RFLP technic to analyse the SNP population genetic characters in Gushi AnkaF2resource population; and associations between SNPs and broiler growth traits,carcass traits, meat quality and serum biochemical parameters were determined.According to the analysis of the genetic variation at the DNA level and the genemutation of the impact on the growth of chicken, we further studied Pax7geneexpression law of different development stages at the mRNA level, and furtherspeculated the relationship between the gene expression and the growth of chicken.The research will provide more clues for the Pax7gene function. The main results areas follows:1. Two SNPs of g.42723T>C, g.3194-3224of the Pax7gene were found in exon5and intron3, respectively. The g.42723T>C was a synonymous substitution and theg.3194-3224is a31bp indel.2. According to the analysis results: The g.42723T>C had significantassociations with chicken body weights at4,8,10and12weeks of age, shank lengthat12weeks of age, chest width at4weeks of age bodyweight, breast bone length at12weeks of age bodyweight, pelvis breadth and body slanting length at8and12weeks of age, carcass weight, semi-evisceration weight, evisceration weight, breastmuscle weight, leg muscle weight, liver weight, heart weight, head weight and clawweight (P<0.05). The g.3194-3224had significant associations with chicken bodyweights at4,6,8,10and12weeks of age, shank length, breast bone length, pelvisbreadth, body slanting length at8weeks old and shank length, shank girth, chestwidth, breast bone length, body slanting length at12weeks of age, carcass weight,semi-evisceration weight, evisceration weight, breast muscle weight, leg muscleweight, liver weight, heart weight, head weight and claw weight(P<0.05).3. Pax7gene were generalized expression in different development stages of theorganizations, and at1day age and36weeks age, glandular stomach, heart, kidneys,chest muscle, the cerebellum, leg muscle and small intestine in higher quantity. At1 day age, liver, glandular stomach, muscular stomach, kidney, hypothalamus, weresignificant higher (P<0.05).4. Pax7gene was highly expressed in leg muscle tissue of individual chicken,and the genotype ID was expressed significant higher than II and DD (P <0.05). TheID and DD were close to significant difference (P=0.056) and II and DD was notsignificant (P>0.05). The same genotype of the cock and the hen were not significant(P>0.05). |
