This research aim to explore the cultivation of the embryonic stem cells (ESC) ofrabbit in vitro and to investigate the healing effect to the soft tissue wound; To provideexperimental basis for the rabbit embryonic stem cells in the application of woundrepair; To investigate the effect of the rabbit ESC secretions on the body woundhealing; To investigate the species specificity of rabbit ESC and the wound repairfunction for different types of trauma; To provide experimental basis for the rabbitESC secretions in the application of wound repair.1Rabbit ESC culture, differentiation, batchesChoose natural estrus female and male rabbit, put them together for the natural mating,after mating90hours, get out of the embryo. Using the enzyme digestion method forprimary culture, observation of cell morphology and change, take to grow goodvaccination on the feeding layer of embryonic stem cells, put in38℃and5%CO2incubator to continue to develop.2The extraction of rabbit embryonic stem cell cultureWhen the ES cells stability batches, start to use pancreatic enzyme digestion method.Now put the nutrient solution together into the centrifuge tube, centrifuge (4000rpm,5min), get supernatant, save frozen.3Build the tissue damage model in the Japanese big ear rabbit.4Build the eye tissue damage model in the Japanese big ear rabbit.5Build the burn model in the Japanese big ear rabbit.6. Build the soft tissue injuries model in the Japanese big ear rabbit.Result:1In the tissue damage models, injection of rabbit embryonic stem cells on the rightside of the wound and no injection of rabbit embryonic stem cells on the left side ofthe comparison, the right side to heal faster. Injection of rabbit ESC on the right side of the wound healing and injection of rabbit ESC culture on the left side of thewounds, the healing without significant difference between the two.2In the eye tissue damage models, the right eye which dripped rabbit ES cell culturesto heal fast.3In the burn models, injection of rabbit ES cell culture on the right side of the woundin comparison with the left side to heal faster.4In the soft tissue injuries models, compared with the two sides, the right side woundof injection of rabbit ES cell culture healing faster and there was no rejection.Conclusion:Through prick transparent zone method and the embryo segmentation method mayeasy make embryo stick wall and value-added, then we can get rabbit embryonic stemcells; Rabbit embryonic stem cells can accelerate wound healing for clinical use;Rabbit ESC culture is easy to collect and convenient to clinical application, it also canstimulating regeneration of tissue cells and inhibiting the apoptosis of damaged cells,good to tissue wound healing; Rabbit ESC culture has no species and no rejection, itis advantageous to the animal wound healing.
